Abstract:
This diploma thesis with the theme of The benefits of green spaces deals with the benefits of green space and their value. The main part of the thesis is a literary review summarizing the theme of the benefits of green space and the methods of their evaluation. The first part of the review focuses mainly on the identification of the functions of green spaces in an urban environment and a closer look at their effects, utility and functioning. In this section the effects of the phenomena that occur in urban environments and which are suppressed thanks to green spaces are also analyzed. The second part of the review is devoted to the importance of evaluating green spaces and their benefits and further to methodologies, which are discussed in detail. The following section analyzes the evaluation methodologies and their positive and critical points are identified. This section also divides the methodologies according to their relevance in the Czech environment, which is specific and the methodologies developed in the Anglo-Saxon context cannot be applied without modification. This chapter also compares data sources needed to develop a methodology for evaluating the benefits of green spaces. In the fifth chapter of the thesis, a methodology for calculating the economic value of the benefits of green spaces in the city is created, based on the knowledge gained in the literature review. The methodology combines methods from various sources and adjusts them to the Czech context. It also examines the data sources needed in order to set up the whole balance sheet. The sixth part deals with the discussion, in which is the summary of the data, which arise from the assembled methodology and compares them with the literature and the application of similar methodologies in other territories. The conclusion summarizes the information obtained in the research and suggests other possibilities of their future use.

Diplom work is describing woody composition of gardens in locality Dobříšsko. There were researched 50 gardens in this locality. Basic dandromatical parameters were found out (height, diameter of trunk, diameter of tree crown), the age of tree was estimated (determined), vitality and health condition of individual wood were rated and then care about greenery vas rated. Basic dandrometrical quantities were got with help of methodics of Agency for nature conservation and landscape Valuation of trees outside forests. The information about care about greenery, about tree felling, about signs of drought in 2015 were found out by questionnaire. Researched data were subdueded basic statistical analysis in program Microsoft Excel.

Abstract:
The thesis Urban green spaces and its institutional framework attempts to analyze different cases of urban green spaces foundation and administration in which various public and private entities act. Based on these foundations it tries to come with an overview of the most important forms of provision of greenery in cities. Through research of available literature we came to the understanding that greenery in urban environments is not just a result of municipal service provided by a public authority but to its foundation, administration, financing and maintenance contribute also other entities. The overview created in the thesis is not exhaustive but can serve as an inspiration for various subjects at a time when local public authorities do not have sufficient resources to ensure satisfactory range and quality of public green.

La presente investigación se centra en los aportes que el poeta chileno Ludwig Zeller ha realizado en torno a la figura femenina como parte de la reivindicación y visualización de la imagen de la mujer. Se revisan en esta tesis, elementos importantes de la escritura del poeta que sitúan a la mujer como pilar fundamental de su obra creativa. Se analiza y compara su propuesta escritural con las tesis de tres teóricas feministas y se interpretan cuatro libros/poemas dedicados a diferentes mujeres de la historia. Finalmente se concluye el aporte artístico de este poeta -situado dentro del movimiento surrealista- y sus implicancias en la literatura contemporánea.

Este trabalho tem como propósito estudar os principais calendários solares desenvolvidos ao longo da civilização humana, incluindo seus mecanismos de construção baseados inicialmente em observações astronômicas até chegar ao calendário atual, em que os movimentos de translação e de rotação do planeta Terra em relação ao Sol, passam a ser substituídos por relógios atômicos. A temática relativa a calendários é altamente interdisciplinar, pois envolve inúmeros aspectos de natureza cultural, histórica, religiosa, filosófica, astronômica e faz uso de conceitos sobre órbitas planetárias, abrangendo ainda a disciplina de física. Por outro lado, conceitos matemáticos, como, divisibilidade, sistemas de numeração e congruências permitem construir o calendário de um ano arbitrário no passado ou no futuro e saber exatamente em que dia da semana cairá uma determinada data. Além disso, considerando que as órbitas planetárias são elípticas, naturalmente comparece o conceito de cônicas. Vale salientar que os Parâmetros Curriculares Nacionais (PCN's) preconizam a exploração de temas e eixos transversais. Inúmeras atividades didáticas integram o escopo desse trabalho e foram propostas para serem exploradas junto aos estudantes do ensino fundamental e médio com o propósito de despertar, motivar e estimular o interesse dos mesmos pelo estudo de conceitos matemáticos e mostrar como a matemática está presente no cotidiano da vida de todas as pessoas, desde as antigas civilizações. Entre as atividades podem ser destacadas, o cálculo do número de dias entre duas datas quaisquer, que é usado para se computar juros bancários e a data de aposentadoria de um trabalhador, assim como a construção de um calendário gregoriano para um ano qualquer a partir de 1582 - data de sua promulgação.

Abstract:
The connection between text, music, and performance in the lieder of the late eighteenth and early nineteenth centuries is an integral aspect to fully comprehending the style and performance of the genre. It is also essential in order to understand the full development of the lied in its totality. The era represented a transitional period in musical development, influenced by Enlightenment values of elegance, good taste, simplicity, and naturalness which sought to eradicate the overly decorative “excesses” of the high-Baroque. In this study, emphasis is placed upon the unique development of the lied in the northern German regions by the composers Johann Friedrich Reichardt and Carl Friedrich Zelter and their musical settings of the lyric poetry of Johann Wolfgang von Goethe. The study also addresses the overall development of the genre as it progressed from the Baroque through Classicism/Neo-Classicism, Sturm und Drang, and into Romanticism exploring the musical settings and performance possibilities both then and now in the context of the various treatises and correspondence between the composers and poet. It seeks to effectively address the notion that these early songs were composed and performed by those versed in the ideal of music being an improvisatory/dramatic vehicle for expressing emotion and textual meaning. In opera, and to a lesser extent other vocal idioms, musico-dramatic excesses occurred in the late Baroque and the cult of the singer reigned. However, the reforms which led to the new aesthetic of naturalness did not suddenly end this improvisatory vocal performance practice. The musical complexity of the lied was gaining in prominence but not yet to the detriment of the priority of the poetic text and its effective rendering.

In den letzten Jahren ergaben sich vermehrt Hinweise, dass dendritische Zellen (DCs) zu einer Stimulation von Natürlichen „Killer“ (NK)-Zellen in der Lage sind, die als zytotoxische Effektorzellen des angeborenen Immunsystems Tumorzellen eliminieren. Aus diesem Grund bestand ein wesentliches Ziel dieser Arbeit in der Analyse der Wechselwirkungen zwischen nativen DCs und NK-Zellen. Dazu wurden slanDCs verwendet, welche die größte DC-Subpopulation des Blutes repräsentieren. Zunächst wurde evaluiert, ob slanDCs eine effiziente Aktivierung von NK-Zellen bewirken. Als ein Ergebnis zeigte sich, dass Lipopolysaccharid (LPS)-stimulierte slanDCs sowohl zu einer verstärkten Expression des Aktivierungsmarkers CD69 auf der Oberfläche von NK-Zellen als auch zur Induktion der NK-Zell-Proliferation führen. Darüber hinaus wurde erstmals die slanDC-abhängige Erhöhung der Expression von aktivierenden Rezeptoren (NKp46, NKp44, NKp30) und Korezeptoren (2B4, DNAM-1) auf NK-Zellen demonstriert, welche essentiell für die NK-Zell-vermittelte Erkennung und Lyse von Tumorzellen sind. In weiteren Untersuchungen induzierten LPS-aktivierte slanDCs eine erhebliche Produktion von Interferon (IFN)-gamma in NK-Zellen, welches proliferationshemmend auf Tumorzellen und aktivierend auf T-Lymphozyten wirkt. Funktionelle Analysen ergaben, dass aktivierte slanDCs das zytotoxische Potential von NK-Zellen gegenüber der Tumorzelllinie K-562 deutlich verstärken. Untersuchungen der zugrunde liegenden Mechanismen zeigten die herausragende Bedeutung von IL-12, das sowohl die Steigerung der IFN-gamma-Sekretion als auch die Zunahme der zytolytischen Aktivität von NK-Zellen induzierte. Darüber hinaus konnte erstmals gezeigt werden, dass LPS-aktivierte slanDCs eine Zytotoxizität von NK-Zellen gegenüber frisch etablierten Blasten von Patienten mit akuter myeloischer Leukämie induzieren. In weiteren Untersuchungen wurde evaluiert, ob NK-Zellen ihrerseits die immunstimulatorischen Eigenschaften von slanDCs beeinflussen. Die Analysen zeigten erstmals, dass unstimulierte NK-Zellen die Expression von MHC-Klasse II-Molekülen, kostimulatorischen Molekülen und Adhäsionsmolekülen auf slanDCs deutlich erhöhen und somit ihre Fähigkeit zur Aktivierung von CD8+ T-Lymphozyten sowie CD4+ T-Helferzellen fördern. NK-Zellen führen ebenfalls zu einer deutlichen Verstärkung der Produktion von IL-12 durch LPS-stimulierte slanDCs. Darüber hinaus zeigte sich, dass NK-Zellen die Sekretion des immunsuppressiven Zytokins IL-10 durch LPS-stimulierte slanDCs reduzieren. In weiteren Analysen wurde demonstriert, dass die Interaktionen mit NK Zellen die Fähigkeit von LPS-aktivierten slanDCs zur Programmierung naiver CD4+ T-Lymphozyten in IFN-gamma-produzierende T-Helfer-1-Zellen deutlich verstärken. Diese Ergebnisse zeigten deutlich, dass stimulierte slanDCs und NK-Zellen in der Lage sind, sich wechselseitig zu aktivieren. NKT-Zellen repräsentieren eine weitere bedeutende Effektorzellpopulation der frühen antitumoralen Immunabwehr, die durch Sekretion von Zytokinen und ein ausgeprägtes zytolytisches Potential zur Elimination von Tumorzellen beiträgt. Deshalb wurden im Rahmen dieser Arbeit erstmals die Wechselwirkungen zwischen slanDCs und NKT-Zellen analysiert. Dabei verstärkten LPS-stimulierte slanDCs die Expression des Aktivierungsmarkers CD69 auf NKT-Zellen. Darüber hinaus induzierten LPS-aktivierte slanDCs eine deutliche IFN-gamma-Produktion in NKT-Zellen, wobei erneut die zentrale Rolle von IL-12 gezeigt wurde. Diese Ergebnisse demonstrierten, dass stimulierte slanDCs zu einer effektiven Aktivierung von NKT Zellen in der Lage sind. In abschließenden Untersuchungen wurde die Wirkung von NKT-Zellen auf slanDCs evaluiert. Dabei verstärkten NKT-Zellen die Maturierung von slanDCs erheblich und führten zu einer signifikanten Steigerung der IL-12-Produktion sowie zu einer Reduktion der IL-10-Freisetzung in Abhängigkeit von IFN-gamma. Die gewonnenen Daten demonstrierten, dass NKT-Zellen und slanDCs zu einer gegenseitigen Aktivierung befähigt sind. Die im Rahmen dieser Dissertation gewonnenen Erkenntnisse zu den Interaktionen von slanDCs und NK- bzw. NKT-Zellen können einen wesentlichen Beitrag zum Verständnis der Immunabwehr von Tumoren leisten und die Konzeption neuer antitumoraler Therapiestrategien unterstützen.

Abstract:
Low-energy X-rays in the range 10 - 50 keV have a wide application. One important application in radiological diagnostics is mammography, whereas, in radiotherapy, they are used for irradiation of superficial tumours, in brachytherapy and photon activation therapy. The importance of soft X-rays for fundamental radiobiological research is based on the fact, that all species of ionizing radiation produce a wide spectrum of secondary electrons, mainly responsible for the primary damage to be transformed into an observable radiobiological effect. By variation of the primary soft X-ray energy, a variation in the secondary electron spectra and therefore in the local energy deposition is provided. However, up to now no definitive conclusions about the relative biological effectiveness (RBE) of soft X-rays can be made due to its dependence on the photon energy, biological endpoint and dose range and the consequent large spread of the published data. The superconducting electron linear accelerator of high brilliance and low emittance (ELBE) at the Forschungszentrum Rossendorf with an electron energy up to about 40 MeV is used, among all, to generate X-rays in a wide energy range. One method for production of intensive, quasi-monochromatic X-rays in the energy range 10 - 100 keV tunable in energy, is by channeling of electrons in a perfect crystal. This X-ray source has many advantages over the most widespread X-ray source, which is the X-ray tube. Although the physical basis of the channeling effect has been previously investigated, the feasibility of an X-ray source based on channeling radiation (CR) for radiobiological studies has been for the first time theoretically and experimentally studied and a dedicated CR source built and optimised in the frame of this thesis. CR has been theoretically characterised in order to estimate its applicability for radiobiological studies. A good agreement between the theoretical predictions and the measured data has been found. The intrinsic properties of the CR source have led to the conclusion that monochromatisation is necessary. A monochromator based on HOPG mosaic crystals, was designed and manufactured. The dosimetrical methods have been investigated at the CR source as well as at an X-ray tube. Absolute dose measurements using an ionisation chamber and spectral dose distribution determination using semiconductor detectors have been performed. In addition, an unconventional system based on thermally stimulated exoelectron emission (TSEE) detectors, allowing to measure dose in a small volume and in the real cell environment has been tested and has proven itself appropriate in a variable dose range and in a liquid environment, in cases where reproducible laboratory conditions are provided. The biological endpoints clonogenic cell survival and micronucleus induction have been optimised for two established cell lines. The human mammary epithelial cells MCF-12A have been chosen due to the importance of RBE of soft X-rays for determination of risk from mammography. On the other hand, the use of the widespread mouse fibroblast cell line NIH/3T3 allows to compare the results with previously published data. The influence of the specific irradiation procedure at ELBE on the control level of cell survival and micronucleus induction has been tested and an irradiation system was developed and constructed. In addition, the RBE for soft X-rays was determined by X-ray tube irradiation at the Medical Department of Technische Universität Dresden. The RBE of 10 kV and 25 kV X-rays relative to 200 kV X-rays was determined. The RBE was found to be in the range from 1.0 to 1.4, depending on the used radiation quality, cell line and the biological endpoint, in agreement with previously published data for the same radiation qualities. These results confirm that systematical studies of RBE dependence on photon energy at the ELBE CR source are necessary and feasible.

Abstract:
Sebbene Anarsia lineatella sia un fitofago ormai da anni introdotto e diffuso in Italia, ancora poco si conosce circa le sue abitudini riproduttive. In considerazione della elevata dannosità di A. lineatella nei pescheti del nord Italia e delle scarse conoscenze sulla sua biologia, si è evidenziata la necessità di approfondire la conoscenza di questo fitofago. Pertanto gli scopi di questa ricerca hanno riguardato l’approfondimento delle conoscenze sui sistemi di comunicazione intraspecifici utilizzati da A. lineatella; la valutazione dell’efficacia di diverse miscele di feromone sintetico, in laboratorio utilizzando anche densità diverse di popolazioni di anarsia e in campo utilizzando gabbie di accoppiamento appositamente costruite, posizionate in frutteti a conduzione biologica nel nord Italia e messe a confronto con gabbie che utilizzavano come fonte attrattiva femmine vergini di tre giorni di età. Sono state condotte prove sul comportamento di maschi di A. lineatella di differenti età in risposta al feromone emesso da femmine vergini di tre giorni di età e al feromone emesso da erogatori di materiale plastico contenenti differenti miscele di feromone sintetico. Sono stati condotti studi per verificare l’influenza del contenuto di alcol ((E)5-10:OH) nella miscela feromonica sulla capacità di inibizione degli accoppiamenti, sottoponendo gli insetti a differenti concentrazioni di feromone in modo da verificare eventuali differenze di attività delle diverse miscele, differenze che emergerebbero con evidenza maggiore alle minori concentrazioni. Alcune prove sono state effettuate anche con differenti densità di popolazione, poiché una maggiore densità di popolazione determina una maggiore probabilità di accoppiamento, evidenziando più chiaramente i limiti di efficacia della miscela utilizzata. Inoltre sono state effettuate prove di campo per confrontare due modelli di erogatore per la confusione sessuale di anarsia contenenti miscele con differenti percentuali di alcol Inoltre, poiché nei pescheti la presenza di A. lineatella è pressoché sempre associata a quella di Cydia molesta e l’applicazione del metodo della confusione deve spesso essere applicato per controllare entrambi gli insetti, può risultare vantaggioso disporre di un unico erogatore contenente entrambi i feromoni; è stato quindi valutato un erogatore contenente una miscela dei due feromoni per verificare eventuali interazioni che possano ridurre l’efficacia.
STUDY OF INTRASPECIFIC COMMUNICATION IN ANARSIA LINEATELLA ZELLER Although Anarsia lineatella is a pest introduced and spread in Italy since many years, little is known about its reproductive behaviour. Considering that A. lineatella causes high damage in northern Italy peach orchards and there is insufficient knowledge on its biology, the need to deepen the knowledge of this pest has emerged. This research has focused on: the study of intraspecific communication systems used by A. lineatella; the evaluation of the effectiveness of different blends of synthetic pheromone also in the presence of different population densities of anarsia; the evaluation of the effectiveness of different blends of synthetic pheromone using mating cages placed in organic orchards in northern Italy. Bioassays were carried out on the behaviour of A. lineatella males of different ages in response to the pheromone emitted by three days old virgin females and to the pheromone emitted by plastic dispensers containing different mixtures of synthetic pheromone. Studies were conducted using some blends with different alcohol content ((E)5-10:OH) in order to evaluate their ability in disrupting the mating. Some bioassays were also carried out with different densities of population, since a greater population density leads to higher probability of mating, to show clearly the efficacy of the mixtures. Field tests were also carried out to compare two mating disruption dispenser models containing mixtures with different percentages of alcohol (for A. lineatella). Since the presence of A. lineatella is often associated with that of Cydia molesta, the application of a single pheromone dispenser for both insects, could be convenient, ensuring that the simultaneous presence of two pheromones does not result in interactions that reduce their effectiveness.

Abstract:
Although adhesion of bacteria and yeast have been extensively studied by a wide range of experimental and theoretical approaches, significantly less attention has been focused on microalgal adhesion to solid materials. Therefore this doctoral thesis is focused on physico-chemical aspects of microalgal adhesion. The results are based on experimental characterization of surface properties of both microalgae and solids by contact angle and zeta potential measurements. These data were subsequently used in modeling the surface interactions (thermodynamic, classical and extended DLVO models) resulting in quantitative prediction of the interaction intensities. Finally, the model predictions are compared with experimental adhesion tests of microalgae onto model solids in order to identify the physico-chemical forces governing the microalgae-solid interaction. The model solids were prepared in order to cover a wide range of properties (hydrophobicity, surface charge). The results revealed that in low ionic strength environment the adhesion was influenced mostly by electrostatic attraction/repulsion between surfaces, while with increasing ionic strength grew the importance of apolar (hydrophobic) interactions. Among tested materials the highest adhesion of Chlorella vulgaris cells was observed to (3-aminopropyl)triethoxysilane modified glass (APTES glass) mimicking the surface properties of stainless steel. The impact of solid surface properties on the degree of colonization by microlagae was statistically more significant than the influence of culture medium composition on cell surface of Chlorella vulgaris.
