Анотація:
The acrylic acid copolymerized with allyl polyethenoxy ether, to produce the dispersant for LiFePO4 lithium ion battery slurry. Taking the quality of acrylic acid as a benchmark, the orthogonal experiment was used to optimalize the burden: the acrylic acid (AA) is 100%, maleic anhydride (MA) is 10%,ethyl acrylate (EA) is 2%, sodium allylsulfonate (ALS) is 6%, allyl polyethenoxy ether (APEG1200) is 60%. The effect of dispersant concentration to slurry fluidity was studied,LiFePO4 content in pulp was 75%, graphitized carbon black was 0.75%,the optimum amount of the dispersant is 0.3wt% of the LiFePO4, the slurry initial viscosity is 1806 mPa.s and 2337 mPa.s after 90min which meet requirements for coating technology.

Diploma III Electrical Technology Study Program is a institutional vocational education institutional in Department of Electrical Engineering and Informatics that aims to produce graduate who are ready to work in operation and maintenance of power system. Since phenomena of scarcity of fossil fuels, study program meet the 2 major problems, namely the limitations of electrical energy for practical lab work and increased job skills on the electrical energy conversion of electrical energy from renewable energy. The purpose of this research is to optimalize capacity of solar and wind energy contained in the environment of the laboratory on the microgrid configuration, namely PV-Wind turbine-Battery. Software HOMER is used to simulate microgrid configuration with AC-DC load, AC load, and DC load. The results show indicate that microgrid PV-Wind turbine-Battery is more economically to meet the need AC-DC load than the others.

Анотація:
This research aims to produce a valid chemistry module based conceptual change model Explore, Diagnose, Design, Discuss and Use (ED3U). The steps of this model are align with scientific approach which is an approach used in current curriculum. This model make the student find their own conception and considering their prior conception will resist or they need alternative conception so that they can have scientific conception. In order to optimalize this, the module is presented using three level chemistry representations that are macroscopic, submicroscopic and symbolic. Developments of this module used 4-D model (define, design, develop, disseminate) with check list as a non test instrument for observation in define step. Beside, it also use questioner to determine whether the module is valid or not. The data was analyzed by Kappa Cohen Formula. Based on the three validators who are the chemistry education lecturers and two validators who are lecturers of Design Communication Visual found that the value of k is 0,829 for content validation and 0,69 for graphics validation. It means the module is valid in high category.

Анотація:
The quality improvement of ramie, including inner and outer aspects are required to be developed to optimalize its production. Unlike horticulture and food crops, ramie cultivation is not constrained by the soil type. This study was aimed to evaluate the agronomical characteristics of three ramie clones in ultisol land. This study was performed using descriptive analysis method with purposive sampling by collecting 10 clumps per clone. Clones of fiber ramie used were Ramindo 1, Bandung A and Lembang A. All clones were planted in ultisol soil located in 350 m above sea level from September 2015 until March 2016. The highest fiber production was achieved from Ramindo 1 followed by Bandung A and Lembang A. The resulted fiber from all clones was considered as quality class II fiber marked by the nominal value ranging from 400-450. Bandung A exhibited the best quality of fiber with nominal value of 450. In terms of its chemical composition, fiber produced by Lembang A contained the highest cellulose (74.7%) with the lowest lignin content (3.6%).

Intisari — Air merupakan sumber kehidupan yang sangat penting bagi kehidupan manusia sebagai bahanbaku air minum, keperluan rumah tangga, dan sumber energi pembangkit listrik tenaga air. Kondisi perairansangat dipengaruhi oleh aktifitas manusia. Salah satu dampak negatif dari aktifitas manusia yaitu timbulnyalimbah yang dapat merusak ekosistem perairan, dengan mengetahui kondisi perairan kita dapatmengoptimalkan penggunaaan air dalam kehidupan. Oleh karena itu, diperlukan sistem yang dapat memantaukondisi suhu dan kadar keasaman (pH) perairan secara real-time menggunakan PC dan nirkabel. Sistem inidinamakan sistem akuisisi data yang menggunakan sensor suhu, modul analog pH meter, telemetri, dan USV.Pemeroses data menggunakan Arduino Mega 2560. Media transmisi data nirkabel menggunakan Telemetri Kit433 MHz. Media penampil data menggunakan PC berbasis GUI (Graphical User Interface) dengan perangkatlunak LabVIEW. Dalam menentukan kondisi perairan, dapat dilakukan dengan mengetahui kondisi beberapaparameternya yaitu parameter fisik (suhu) dan parameter kimir (pH). Berdasarkan hal tersebut dilakukanpengukuran nilai suhu dan pH pada perairan. Sistem akuisisi data yang dilengkapi dengan sensor suhu DB10S20, Analog pH meter, Arduino Mega 2560, dan Telemetri Kit 433 Mhz. Sistem ini diletakan pada USV yangtelah diatur untuk bergerak menuju titik waypoint, melakukan pengukuran pada titik waypoint tersebut danmenampilkan data serta menyimpan data dengan data logger. Sensor DB 10S20 dikalibrasi menggunakanFLUKE 62 MAX IR Thermometer dan Analog pH meter menggunakan pH indikator universal. Sistem ini dapatdigunakan selama sumber tegangan mencukupi serta jarak pengiriman data yaitu line of sight ≤ 50 m dandalam bangunan ≤ 15 m. But there are some negative impact that produced by human activity that coulddamages the water as important life resource. Yet, by analyzing the water condition, we may optimalize theusage of water. Then, it is necessery to have a proper system which able to supervise the water temperature andacid content (pH) in real-time by using PC and wireless equipment. This system is called by data acqusitionsystem which include the usage of temperature sensor, an analogue modul pH meter, telemetry and USV.Arduino Mega 2560 is chosen as data processor in this research case. Wireless transmission data isacommodated by Telemetry Kit 433 MHz. While data performance media is provided by GUI (Graphical UserInterface) based PC as specified software known as LabVIEW. There are some parameters to be connected tothe water condition, which are physical parameter (temperature) and chemical parameter (pH). Based onmethod above water temperature and pH is measured. Data acquisition system is supported by DB 10S20temperature sensor, pH meter analogue, Arduino Mega 2560 and Telemetry Kit 433 Mhz. This system is placedon USV which systemized to move forward to the waypoint spot, then it start to measure at the waypoint spot,perform and save the data by data logger. DB 10S20 sensor is calibrated by FLUKE 62 MAX IR Thermometerand the pH meter analogue is using pH universal indicator. The system could be occupied as long as the powerresource is able to covered the data delivering in line of sight of ≤ 50 m and ≤ 15 m indoor. Blimbingsari beach is one of a kind of Banyuwangi’s beach that have a high potential on grilled fish special culinary aspect This beach became the pioneer of Banyuwangi’s very first seafood and grilled fish restaurant with ‘lesehan’ type, and that was the brand was formed ever since. Thus also makes the image of Blimbingsari skyrocket and built perspective of tourists for years and the paradigm brings many people to come and try the dish by themselves. However, Blimbingsari beach has not optimalize the brand identity they have and has no tagline or destination icon to increase the branding yet. The main purpose of this research is to formulate of priority strategies of Blimbingsari destination branding as a grilled fish culinary center which lies in Banyuwangi.This research is using qualitative descriptive with SWOT analysis and AHP for making desicion in a path of top priority strategies. And the result itself, shows that the culinary potential has been being built by brand and long-journey tourist perspective, the taste of the grilled fish itself, best price dishes and comfortable ambiance. The highest strenght factor owned Blimbingsari beach is having a strategic location, the main weakness factor is inadequate food utensils. The major opportunity of factor is the sinergy between to communities to increase regional profit and the main threat factor is the emergence of new competitors. In addition, this research also produced five priority strategies.

Анотація:
This diploma thesis focuses on the optimization of bacterial cellulose production by Komagateibacter xylinus DSM 46604. The theoretical part of this thesis describes the properties of bacterial cellulose, its production and application possibilities. The experimental part aimed to assess the effect of different cultivation conditions on the production yields of bacterial cellulose. The effects of several cultivation strategies have been studied such as: (1) effect of acetate buffer used as a medium, (2) impact of oil added into the medium, (3) fed-batch cultivation, (4) variation of the volume of cultivation vessel and cultivation media and (5) aeration. In addition to the production of relatively thin samples, up to 100 µm was synthesized unique 3D structured bacterial cellulose in the form of cylinders, with a height up to 2 cm. The growth of bacterial cellulose in the form of cylinders was achieved by dynamically cultivating K. xylinus in combination with fed-batch approach.

In this thesis, optimization of production of lipolytic enzymes by submerzed cultivation of Bacillus subtilis (BS) was studied. Production of lipolytic enzymes was tested in three nutrient media, which differed mainly in main sources of carbon, respectively of nitrogen. The first medium contained mainly extract from calf brain and beef heart (BHIB), the second medium contained peptone and yeast extract (NB) and the third one contained peptone and yeast extract with the addition of 2% (w/v) glucose (NBG). The highest lipolytic activity (0.0784 Uml-1) was measured in NBG medium. Maximum of lipolytic activity was observed before the end of the exponential phase of BS growth in all the media. Temperature optimum in NBG medium was determined from 30 to 50 °C, pH optimum in the range of 5 to 11 and subsequently the temperature stability of lipolytic enzymes produced by the BS was estimated. The activity value was determined spectrometrically using p-nitrophenyllaurate as a substrate. Produced lipolytic enzymes showed maximum activity at 37 °C in the alkaline pH of 8.0. Measurement of temperature stability showed that lipolytic enzymes are relatively thermostable enzymes retaining 100 % of the activity even after 1 hour of cultivation at 30 - 50 °C. The presence of 1% (w/v) olive oil in medium NBG caused a decrease in lipolytic activity by 65 % as well as in pH from 6.5 to 5.4 after 14 days of cultivation. After substitution of glucose by fructose in medium NBG, lipolytic activity showed comparable values during the first week of cultivation. On the other hand, the decrease of lipolytic activity by 29 % in the medium with fructose was observed after 14 days of cultivation. A procedure for the identification of lipolytic enzymes of BS by peptide massfingerprinting was developed to understand the potential of synthetic polyester - poly(e-caprolactone) as a lipase inductor. Degradation study of commercial polyester poly(e-caprolactone) was carried out by submerged cultivation of Bacillus subtilis in NBG medium at initial pH 7.0 and 30 °C for 14 days. PCL (Mn = 10,000, Mw = 14 000) was studied in the form of films (1.0 x 1.0 cm), which were prepared by melt-pressing, rapid cooling of the melt to 4 °C and evaporation of the solvent from 2 % dichlormethane solution. The evaluation of the films shown occurrence of weight loss (7.8 - 17.0 wt.%) together with the formation of numerous holes and cracks in the sample surface in relation to the method of the films preparation. Lipolytic activity values increased by 9 - 17 % in the degradation media compared to control samples. Densitometric monitoring showed also higher increase in cell mass in the degradation medium compared with control samples. Based on the results obtained, the degradation process induced by BS could be suggested.

V této diplomové práci je popsána přechodná transfekce buněk 293 HEK adaptovaných na růst při suspenzní kultivaci bez přítomnosti séra za použití polyethyleniminů (PEI). Buňky byly transfekovány plasmidem pcDNA5/SEAP, který exprimuje sekretovanou formu lidské placentální alkalické fosfatázy. K porovnání účinnosti jednotlivých transfekcí byla měřena koncentrace exprimované fosfatázy v buněčném supernatantu. Cílem této práce bylo optimalizovat různé faktory ovlivňující účinnost transfekcí s důrazem na nalezení optimálního poměru DNA:PEI.

Diploma thesis deals with use of Zymomonas mobilis for the production of bioethanol from waste paper. There were used three kinds of substrate (cardboard, drawing and office paper) to optimize of bioethanol production. Individual papers were subjected to the same pre treatment, namely a milling, a combination of microwave irradiation and NaOH, a combination of microwave irradiation and H2SO4 and combination microwave irradiation, H2SO4 and NaOH. The substrates were decomposed by enzymatic hydrolysis after pre treatment to evaluate the best pre-treatment. Simultaneous saccharification and fermentation was carried out for each substrate (with two of the best pre-treatment). The samples were taken during the hydrolysis and the simultaneous saccharification and fermentation, and were determined by HPLC. Growth curves of Zymomonas mobilis were constructed, as the most appropriate for SSF was chosen temperature of 40 ° C in which the exponential phase took place at the time of 6 15 hours. During hydrolysis was monitored glucose concentration in the solution. The maximum concentration of glucose was in the cardboard (microwaves + H2SO4 + NaOH) 16.46 gdm-3, a drawing (microwaves + H2SO4 + NaOH) 31.78 gdm-3, and office paper (microwaves + H2SO4) 25.04 gdm-3. The concentration of ethanol for SSF was highest in the same cases as in the hydrolysis. The cardboard was the maximum concentration of bio ethanol 9.5 gdm-3, for the drawing 16.1 gdm-3 and for the office paper 12.13 gdm-3.

In the diploma thesis are discussed the process of enzymatic hydrolysis of waste paper as a source for the production of bioethanol by bacteria Zymomonas mobilis. In the theoretical part summarize basic information about particular methods of hydrolysis, about paper used as a raw material for enzymatic hydrolysis, about possibilities of the fermentative production of bioethanol focusing on the method of simultaneous saccharification and fermentation comparison with enzymatic hydrolysis and fermentation. Suitable microorganisms for ethanolic fermentation and simultaneous saccharification and fermentation and their advantages and disadvantages, are further discussed in this part as well. The theoretical part ends with the suggestion of the technological process for production of bioetanol. It covers all necessary steps from the input of raw material to the separation of produced ethanol. In the experimental part various parameters of hydrolysis, fermentation and simultaneous saccharification and fermentation were optimized using enzymes from Novozymes® company and the Zymomonas mobilis CCM2770 and Zymomonas mobilis LMG457 bacterium. The conversion rate of paper cellulose to gluckose and production of ethanol were observed by HPLC/RI method. Type of buffer, quantity of cells, enzyme and substrate were optimized in order to maximize the efficiency of the process. All experiments were performed on paper containing high amount of cellulose and for comparison on standard medium which contains gluckose. The highest yields was achieved with the use of Novozymes® Cellulosic ethanol enzyme Kit. The strain Zymomonas mobilis LMG457 has demonstrated as a better producer.

Анотація:
Several yeast strains and microalgae were selected for this diploma thesis. -glucans, lipids, carotenoids, ergosterol and coenzyme Q were determined in selected producers, and the cultivation conditions for yeast strains were optimized to gain enhanced production of -glucans. Microalgae cultivations were carried out according to the instructions of the Collection of Autotrophic organisms (CCALA). Selected microalge strains include Desmodesmus acutus, Dunaliella salina, Arthrospira maxima and Cyanothece sp. Selected yeast species include Rhodotorula glutinis, Cystofilobasidium macerans and Sporidiobolus metaroseus. Edible yeast Saccharomyces cerevisiae was cultivated to compare with other yeast strains because of it's verified production of -glucans. -glucans were then determined by the enzymatic kit K-YBGL Megazyme, carotenoids, ergosterol and coenzyme Q were analyzed by HPLC/PDA and fatty acids were analyzed by GC/FID. The best producer of yeast -glucans was R. glutinis and S. metaroseus, and the best conditions for the production of -glucans and other metabolites was the C/N ratio of 70. Within the microalgae species, only -glucan production was observed, the best producer was D. acutus.

The objective of this thesis is to present the basics of production scheduling problems for workstations and to create an application of dynamic model. This application should help the UNIKOV company to produce similar schedules for their own operations. The theoretical part provides a brief description of terms and characteristics of the scheduling problems. Detailed characteristics are provided on the open shop problem - the task applied on the real world example. The paper then follows with a practical part, covering an explanation of the heuristics model written in VBA for MS Excel, allowing dynamic expansion and management of the production.

This diploma thesis deals with the risk associated with fish production. Selected risks are further taken into account. The risks are further described and addressed using the FMEA matrix. The thesis contains a mathematical model that solves the optimal population of the pond in order to maximize profits. The model includes fixed input parameters, which includes the price of hatchery material and the market price of fish. Furthermore, the model contains random input parameters that are limited by the intervals and includes the growth coefficient and the size of the population in the pond. The optimization model is based on simulated data, which are based on real data and statistically determined estimates. The used data are processed into tables in MS Excel for better visualization. GAMS software is used for software implementation of the model.

This thesis deals with the optimalization of production of the thermostat bulb K22 in Invensys Appliance Šternberk Ltd. company. It analyzes the manufacturing process and proposes its optimal solutions. This thesis also describes other possible solutions that contribute to efficient management of the lean enterprise. The main benefit of this thesis is in the reduction of required labor time for the manufacture of the thermostat and in the introduction of lean management of selected tools.

Анотація:
The Diploma Thesis is focused on the problematic of the optimization and consecutive communication of the mobile application which is developed by one Prague shopping center for its needs. For that I will use suitable communication mix based on current experience since the launch of the app which was in September 2017. The communication of the application itself to all interest groups in managed by marketing agency in cooperation with marketing management of the certain shopping center. Though it seems on the first sight that it is mainly B-2-B market, main interest focus is on the final customer satisfaction and that is B-2-C market. There is a detailed analysis of the Prague shopping centers market with emphasis on another mobile applications of the same purpose made in the Thesis. Then there are the steps how to make the marketing communication of the application more effective a so make the awareness of the application better suggested from the results of the analytical part .
