Uncontrolled oxygen is often administered to breathless patients regardless of whether hypoxaemia is present. In acute exacerbations of Chronic Obstructive Pulmonary Disease (AECOPD) this may result in carbon dioxide (CO2) retention and worsening respiratory failure in some patients. In AECOPD the main mechanism is the release of hypoxic pulmonary vasoconstriction and an increase in the physiological dead space to tidal volume ratio (VD/VT). Acute asthma and pneumonia have features in common with AECOPD, namely significant ventilation – perfusion mismatch; and there is the potential for CO2 re

We have established a murine model of pneumococcal pneumonia in order to characterise the host inflammatory response. Intranasal infection with Streptococcus pneumoniae resulted in constant bacterial loads within airways of susceptible mouse strains whilst the pathogen burden increased in both the lung tissues and bloodstream. The bacteria induced inflammation that was evident as perivascular cell recruitment in histological sections. Mast cell granule staining indicated that this population degranulated early in the inflammatory response, releasing TNFa into the lung environment. Levels of TN

Abstract:
Neutrophils are innate immune cells that form part of the first line of defense against pathogens. These cells are rapidly mobilised in response to infection and inflammation and are equipped with a range of pathogen destruction mechanisms. In recent years it has been demonstrated that neutrophils can also modulate innate and adaptive immune responses. Neutrophils have been found to enter secondary lymphoid organs including lymph nodes, however the phenotype and function of neutrophils in lymph nodes is not well understood. Work in this thesis utilised an acute pulmonary infection model with t

The reason of infection leading to ACS maybe explained by increased inflammation, hemoconcentration, imbalance between oxygen demand and supply. Of all bacterial infections, pneumonia is mostly associated with the development of ACS.Streptococcus pneumonia is the most common causative pathogen of community acquired pneumonia, and Streptococcus pneumonia infection is asscociated with clinically severe form of pneumonia. However, relatively few studies

Abstract:
Pneumonia and sepsis are distinct but linked public health concerns. Each condition is the leading cause of the other; however, the responses controlling the susceptibility between the two disease processes remain speculative. The acute phase response (APR) is an important component of the host immune response during pneumonia and sepsis, and primarily driven by the activation of hepatocyte transcription factors NF-κB RelA and STAT3. While the NF-κB pathway is essential for inflammation and hepatocyte function, its inactivation has been associated with hepatotoxicity. Liver injury is an ind
