A series of three experiments were conducted to determine the diversity of indigenous chlorobenzoate (CB) degraders in soil and to investigate the use of different methods of bioaugmentation for remediation of contaminated soil. In the first study, soil was amended with either 500 or 1000 μg of 3-CB g⁻¹ and was either uninoculated or inoculated with the 3-CB degrader Comamonas testosteroni BR60. Bioaugmentation with C. testosteroni BR60 increased 3-CB degradation at both contaminant levels, and the increase was more pronounced at the higher level due to contaminant inhibition of indigenous 3-C

The biological and molecular properties of Sinaloa tomato leaf curl virus (STLCV) were investigated to test the hypothesis that STLCV is a previously uncharacterized whitefly-transmitted geminivirus from North America. STLCV causes leaf curling and yellowing in tomato plants. STLCV was transmissible to N. benthamiana by sap inoculation, and to Solanaceous and Malvaceous species by the whitefly vector. STLCV has transmission characteristics like other persistent viruses, and was not transovarially passaged. PCR fragments containing the large intergenic region (IR) of the STLCV A and B component

Molecularly imprinted polymers (MIPs) are biomimetic sensing elements that combine the accuracy and highly specific binding affinity of a biosensor, with the robustness and reusability associated with artificial electrochemical sensors. This thesis investigates the application of a MIP sensor to address the challenge of phosphate detection in precision agriculture. Traditional chemical sensing approaches using portable electrochemical sensors display a significant cross-interference between inorganic phosphate and other nutrient ions. This is due to the low position of phosphate in the Hofmeis

Blue oat mite species, Penthaleus spp. (Acari: Penthaleidae), and the redlegged earth mite, Halotydeus destructor (Tucker) (Acari: Penthaleidae), are some of the most serious and widespread agricultural pests in southern Australia. Management of these mites has been complicated by the recent discovery of three cryptic species of Penthaleus, whereas prior research had assumed a single species. The species differ markedly in their distribution, plant preferences and response to pesticides. Wheat (Triticum aestivum) kernel texture (hardness) is the most important determinant of milling and end-product quality. Recent data indicate that the only difference between soft and hard textured wheat is a single amino acid mutation in one protein, puroindoline-b (PIN-b). A rare tryptophan-rich domain in this protein consists of five tryptophan residues among a stretch of seven amino acid residues. To understand how this single mutation makes hard wheat possible, thus enabling bread making, it is crucial to have a high-resolution three-dimensional structure of this protein. The prerequisit

Plant mitochondria possess a gene expression system in which post-transcriptional events, such as transcript end maturation and turnover mechanisms play a key role in regulating the transcriptome. The fungus Nectria haematococca is a broad host range pathogen. Isolates pathogenic on pea are able to detoxify the phytoalexin pisatin using the enzyme pisatin demethylase. When the gene (PDA1 ) encoding this enzyme was mutated via gene disruption, the mutants were less virulent but still pathogenic on pea. Additional studies demonstrated that PDA1 was on a 1.6-Mb conditionally dispensable (CD) chromosome and that loss of this CD chromosome resulted in the complete loss of pea pathogenicity. Magnaporthe grisea is a filamentous ascomycete fungus that causes blast disease on rice and other grasses. Understanding the molecular basis of plant resistance is the foundation for the development of unique and durable plant protection.
