Abstract:
Neonatal hyperoxia impairs vascular and alveolar growth in mice and decreases endothelial progenitor cells. To determine the role of bone marrow-derived cells in restoration of neonatal lung structure after injury, we studied a novel bone marrow myeloid progenitor cell population from Tie2-green fluorescent protein (GFP) transgenic mice (bone marrow-derived angiogenic cells; BMDAC). We hypothesized that treatment with BMDAC would restore normal lung structure in infant mice during recovery from neonatal hyperoxia. Neonatal mice (1-day-old) were exposed to 80% oxygen for 10 days. Abstract A major obstacle to using bone marrow cell-based therapies for ischemic cardiovascular disease is that transplanted cells must survive in an ischemic microenvironment characterized by low oxygen, glucose, and pH. We demonstrate that treatment of bone marrow-derived angiogenic cells (BMDACs) with dimethyloxalylglycine, an α-ketoglutarate antagonist that induces hypoxia-inducible factor 1 (HIF-1) activity, results in metabolic reprogramming of these cells, with increased glucose uptake, decreased O2 consumption, increased lactate production, decreased reactive oxygen species, and increa

Abstract The purpose of this study is to examine the influence of bone marrow-derived adipocytes (BMDAs) on the proliferation and metastasis of high-grade serous carcinoma (HGSC), the most common and aggressive type of ovarian and fallopian tube cancer. BMDAs begin as myeloid cells that traffic to adipose tissue and differentiate into adipocytes. While histologically identical to conventional mesenchymal adipocytes (CMAs), BMDAs are a highly inflammatory subtype. BMDAs accumulate within visceral adipose depots and their production increases with age. BMDA production increases in mouse “pseudom

Abstract Introduction: The impact of anesthetic exposure on the immune system is relatively unknown at both mechanistic and clinical levels. In this in vitro study, we employ immunological assays and understanding of immune cell function to evaluate the effect of volatile anesthetic (VA) exposure on bone marrow dendritic cells (BMDCs). BMDCc play a critical role in antigen presentation and link the innate and adaptive arms of the immune response. Hypothesis: We believe that VA exposure may modulate immune responses, specifically BMDC function. Methods: Bone marrow cells were harvested from C57

The aim of this paper was to assess whether the BMDC after freezing and thawing are capable to retain the immunophenotype and antigen-presenting capacity. BMDC were generated from bone marrow precursor cells as described previously by culturing the cells in medium containing GM-CSF and IL-4. Afterwards, the cells were harvested, counted and used for phenotyping and priming of syngeneic spleen cells. For cryopreservation, the BMDC were frozen in the presence of 10% of dimethylsulphoxide (DMSO) and 90% foetal calf serum. Forty to fifty percent of both samples, frozen/thawed as well as fresh BMDC

During the progression of acute renal failure (ARF), the renal tubular S3 segment is sensitive to ischemic stresses. For reversing tubular damage, resident tubular cells proliferate, and bone marrow-derived cells (BMDC) can be engrafted into injured tubules. However, how resident epithelium or BMDC are involved in tubular repair remains unknown. Using a mouse model of ARF, we examined whether hepatocyte growth factor (HGF) regulates a balance of resident cell proliferation and BMDC recruitment. Within 48 h post-renal ischemia, tubular destruction became evident, followed by two-waved regenerat

Abstract Angiogenesis alleviates hypoxic stress in ischemic tissues or during tumor progression. In addition to endothelial cell proliferation and migration, the angiogenic process requires bone marrow–derived cell (BMDC) recruitment to sites of neovascularization. However, the mechanism of communication between hypoxic tissues and the BM remains unknown. Using 2 models of hypoxia-induced angiogenesis (ischemic hindlimb surgery and subcutaneous tumor growth), we show that platelet infusion promotes BMDC mobilization into the circulation, BMDC recruitment into growing neovasculature, tumor vasc

El título obtenido en iBMDC fue significantemente mayor que en mBMDC (12 y 24hpi). Para los aislados 3249 y 3262, la replicación alcanzó el pico antes en las iBMDC (24h) que en las mBMDC (48h). Bangladesh’s Barind Tract, lying in the country’s northwestern part, is a drought-prone water scarce area that has suffered substantial difficulties in water management for agriculture, drinking, residential, and other uses. The situation has been changed by the efforts of the Barind Multipurpose Development Authority (BMDA). So, the focus of this research is on the numerous initiatives of the BMDA to transform the arid-like Barind area into a green and granary landscape through efficient water management.
