I have investigated the possibility that specific conalbumin (ovotransferrin) iron saturation levels enable less virulent strains of Salmonella to become more virulent. Iron starved cells of two pathogenic Salmonella strains, S. paratyphi B var. java and S. thompson, were cultured in iron limited media at 3 different iron conalbumin saturation levels. Results indicate that strains differ significantly at both low and high iron saturation conalbumin. In this thesis, comparative genomic analysis was used to detect variation within the Typhi and Paratyphi A populations, and to compare the evolution of these two pathogens. There was evidence in both populations of ongoing accumulation of inactivating mutations which result in loss of gene function. The O—antigens of Salmonella serogroups A, B and D differ structurally in their side-chain sugar residue. The genes encoding 0antigen biosynthesis are clustered in the rfb operon. This thesis describes the molecular cloning and analysis of the rfb genes from S. paratyphi A IMVSl316 (serogroup A) and S. typhi Ty21a (serogroup D). The rfb gene cluster from S. Typhimurium LT2 (serogroup B) has also been studied in our laboratory (Brahmbhatt et al 1986, 1988; P. Wyk, Ph.D. thesis, Univ. of Adelaide, 1988).
