Drew, Gail L. "DNA-Protein Cross-Linking by Pyrrolizidine Alkaloids." DigitalCommons@USU, 1997. https://digitalcommons.usu.edu/etd/3919.

Abstract:
Pyrrolizidine alkaloids (PAs) are natural plant compounds found in hundreds of plant species worldwide and are reported to have cytotoxic, carcinogenic, antimitotic, and gentotoxic activity. PAs are metabolized by the cytochrome P450 (CYP) sytem to the pyrrole or the N-oxide form. They pyrroles are bifunctional electrophillic alkylators that bind cellular nucleophiles such as DNA and proteins and disrupt normal cell processes, including DNA replication and gene transcription, and can cause megalocytosis. The pyrroles dehyrosenecionine (DHSN) and dehydromoncrotaline (DHMO) are among the most po

Abstract:
Protein-nucleic acid interactions play essential roles in all living cells in various cellular functions. The study of these interactions can reveal important structural and functional information. UV cross-linking of nucleic acids to proteins in combination with mass spectrometry is a powerful technique to identify proteins, peptides and the amino acids involved in intermolecular interactions within nucleic acid-protein complexes. However, the mass spectrometric identification of cross-linked nucleic acid-protein heteroconjugates in complex mixtures and MS/MS characterisation of the specific

Abstract:
The pyrrolo[2,1-c][1,4]benzodiazepines (PBDs) are a diverse group of antitumour antibiotics in which there has been growing interest over the past 30 years. These compounds bind covalently within the minor groove of DNA via the formation of an aminal bond between the N10-C11 imine functionality of the PBD and the exocyclic C2-NH2 of a guanine base, resulting in the inhibition of processes including DNA replication and transcription, thus leading to cell death. PBD `dimers' have the ability to form irreversible interstrand cross-links with DNA. One example, DSB-120, has been shown to recognise

Transcription factors play crucial regulatory role within the cell and the entire multicellular organism. The important factor is its ability to interact with other regulatory proteins and DNA. Despite the fact that a large part of the interaction network is already documented, detailed information on the structure and dynamics of protein-protein and protein-DNA complexes is still scarce. In this thesis we focused on the possibility of studying conformational changes given by the transcription factor-DNA complex formation using the methods of structural mass spectrometry: hydrogen/deuterium ex
