A number of methods are currently used for the detection of chemically-induced chromosome damage but all are subject to the criticisms that they are either time-consuming and labour intensive or require a large number of animals and can be insensitive. However, for human risk-assessment, conclusive evidence that compounds possess potential heritable hazards can still only come from mammalian germ cell studies. Lipotropes (methionine, choline, folate, and vitamin B12) play a key role in one-carbon metabolism. We hypothesized that maternal methyl (lipotropic) nutrients would induce epigenetic modification that influences mammary development and carcinogenesis from dam to offspring (F1 & F2). Prostaglandin E2 (PGE2), a byproduct of arachidonic acid metabolism, has been suspected to be involved in tumor promotion. It has been suggested that diet may modulate PGE2 level in organisms thus affecting the implantation and growth of the tumor tissue. PGE2 content was investigated in mice fed ad libitum four types of fatty acid diets: saturated fatty acid diets: a stearic acid and a palmitic acid, and polyunsaturated fatty acid diets: a low fat (safflower 1%) and a high fat diet (safflower 15%). The ability of mammary tissue from prepubertal bulls and heifers of beef and dairy breeds to respond to hormonal stimuli through synthesis and secretion of milk proteins was studied. The female sex hormone 17beta-estradiol, produced naturally in the body, seems to play an important role in the development of breast cancer, since (i) it can be activated to reactive metabolites, which are known to damage DNA and (ii) the stimulation of the estrogen receptor alpha by 17beta-estradiol enhances cell proliferation.
