A range of low-valent iridium and platinum fluoro-complexes have been prepared and characterized by a combination of mass spectrometry, 19F 31P{1H}, 195Pt{1H} and infrared spectroscopies and EXAFS where possible. [Dichlorobis(amine)platinum(II)] complexes of the type [PtCl2(L)2] (L = NMe3, NH3, py; L2 = en, bipy) have been reacted with [XeF2], undergoing oxidative fluorination, to afford various isomers of platinum(IV) amine-fluoride species. The prevalence of fluorine in natural products is scarce. There are but a handful of compounds that have been discovered to date. Transition metal carbonyls (Ir4(CO)12, Os3(CO)12 and M(CO)3 (PPh3)2 M=Ru or Os) have been reacted with XeF2, and Os3(CO)12 with fluorine gas. The products have been fully characterised by NMR spectroscopy, 19F, 13C, 13C{19F} and 31P where appropriate. Oligonucleotides containing the modification 2-fluoro-2-deoxy-beta-D-arabinose (2'F-ANA) were synthesized via solid phase synthesis in order to investigate their ability to bind to nucleic acids (DNA and RNA). The ability of this modification to serve as a suitable antisense oligonucleotide construct was also evaluated.<br>All four nucleobase monomers (thymine, cytosine, adenine and guanine) were prepared for solid phase oligonucleotide synthesis, from which homopolymeric and heteropolymeric base sequences were assembled. Chapter 1 gives an overview of the fluorine chemistry field, from its early developments to recent applications in medicinal chemistry. The development of asymmetric electrophilic or nucleophilic installation of fluorine in organic molecules is highlighten. Chapter 2 of this thesis discusses the enantioselective synthesis of α-fluoroamides. The study is applied to the synthesis of fluoroenantiomers of the bioactive molecule capsaicin and short-chain analogues.
