Abstract:
Phosphofructokinase (ATP: D-fructose 6-phosphate 1-transferase, EC 2.7.1.11) catalyses the phosphorylation of fructose 6-phosphate to fructose 1,6-bisphosphate, and is a key regulatory enzyme in glycolysis. Sequence conservation between mammalian PFK and PFK from E.coli, B.stearothermophilus and S.citri, and between the amino and carboxyl halves of the mammalian enzymes suggests that mammalian PFK has evolved from a prokaryotic progenitor by a process of gene duplication and fusion. Abstract:
Platelet-activating factor (PAF) is a broadly bioactive family of phospholipids which contribute to the pathogenesis of numerous diseases as well as to many normal physiologic processes. The pleiotropic nature of PAFs actions may be due to the activation of several intracellular signaling pathways or the presence of PAF receptor subtypes. Therefore, to begin to understand the complex mechanisms by which PAF molecules induce cellular responses, a molecular approach was initiated to provide tools to investigate many of the issues surrounding PAF receptors. Neonatal alloimmune thrombocytopenia (NAIT) is caused by maternal alloantibodies against fetal platelet antigens inherited from the father and which are absent from maternal platelets. In Caucasians, antibodies against the Leu33 (HPA-1a) polymorphism of integrin β3 (part of the platelet αIIbβ3 complex) account for >70% of cases. Antenatal screening for these antibodies does not currently take place in the UK, partly because of the absence of sensitive, predictive tests. Abstract:
Previous work on the binding of resting platelets to the basement membrane glycoprotein, laminin, has identified the Ic/IIa integrin c01aplex (CD49f/CD29), also known as VLA-6, as the receptor. There exists however, another protein with a molecular weight of 67kDa, that mediates this function on other cells. It is abundantly expressed on the membranes of breast cancer cells, where it plays a key role in both the localisation at, and penetration of vascular beds, by metastases. Redox balance is important for platelet physiology, indeed it is crucial for blood coagulation and thrombosis. This thesis will give an update on intracellular and plasma membrane systems involved on redox homeostasis in human platelets, with particular regard to vitamin C and plasma membrane NAD(P)H oxidases. The redox balance is linked to the presence of intracellular antioxidants; in particular, vitamin C appears to be a key modulator of platelet oxidative state, since these cells physiologically accumulate ascorbic acid. The sulfhydryl group is considered the major target of HNO. It can be modified into either sulfinamide or disulfide.
