Abstract:
The application of MIDA boronates (MIDA = N-methyliminodiacetic acid) in Suzuki- Miyaura reactions has increased over the last years. This is mainly because in many cases, the replacement of the boronic acid for the respective MIDA boronate has a positive result in the reaction yield. The key feature that makes MIDA boronates efficient coupling partners is that they can undergo a slow hydrolysis reaction under Suzuki-Miyaura conditions, which generates the boronic acid in situ. The control of the hydrolysis rate is crucial to keep a low concentration of the boronic acid to avoid some of the si

Phosphotriesterase (PTE) is a zinc metalloenzyme that catalyzes the hydrolysis of organophosphorus compounds. Metal ion roles during binding and catalysis are probed by comparing the kinetic properties of Zn/Zn, Cd/Cd, and Zn/Cd PTE with a variety of phosphate trisesters. The metal in the α-site of the binuclear metal center modulates the pKa values determined from pH-rate profiles. These results suggest that the α-metal is responsible for activating the nucleophilic hydroxide. In an effort to determine the function of the β-metal, the kinetic parameters for diethyl p-chloro

This cycle has to be repeated eight consecutive times to reduce one molecule of N2. The two catalytic halves of the MoFe protein had been considered to be independent of each other. Tese de Doutoramento em Ciências Veterinárias<br>Xyloglucan (XG) is the major plant cell wall hemicellulose. Biochemical and structural studies on a xyloglucanase, Xgh74A, from Clostridium thermocellum are presented here. In addition, the carbohydrate-binding modules (CBMs) from C. thermocellum Cel9D-Cel44A, CtCBM30 and CtCBM44, which recognize XG, are characterized in this work. CtCel9D-Cel44A’s C-terminal module of unknown function is a CBM, constituting the founder member of family 44. Structural studies revealed that both CtCBM30 and CtCBM44 present Type B binding site topologies whe

Abstract:
A series of lab-scaled digestion studies including conventional mesophilic anaerobic digestion(MAD), thermophilic anaerobic digestion (TAD) at a range of treatment temperatures, and mesophilic high solids digestion of thermally pretreated wastewater sludge (THD) were carried out. Enhanced digestion performance in terms of solids destruction and methane generation by THD relative to MAD was achieved, and was largely attributable to the solubilization and subsequent biodegradation of energy-rich substrates within blended primary and secondary sludge. TAD was observed to underperform MAD, especia

Abstract:
Anaerobic digestion has been the preferred method for reducing and stabilizing waste sludge from biological wastewater treatment for over a century; however, as sludge volumes and disposal costs increase, there has been a desire to develop various methods for reducing the volume of sludge to be treated, improving the performance of the digesters, and increasing the energy value of the sludge. To this end, there have been numerous pretreatment and side-stream systems studied and developed over the past several decades with the overall goal of reducing the volume of biosolids to be disposed of
