Abstract:
Vascular colonisation by Bartonella henselae may cause vaso-proliferative tumour growth with clumps of bacteria found in close association with proliferating endothelial cells. By using B. henselae-infected human umbilical vein endothelial cells as an in vitro model for endothelial colonisation, we report here on a novel mechanism of cellular invasion by bacteria. First, the leading lamella of endothelial cells establishes cellular contact to sedimented bacteria and mediates bacterial aggregation by rearward transport on the cell surface. Subsequently, the formed bacterial aggregate is engulfed and internalised by a unique host cellular structure, the invasome. Completion of this sequence of events requires 24 hours. Cortical F-actin, intercellular adhesion molecule-1 and phosphotyrosine are highly enriched in the membrane protrusions entrapping the bacterial aggregate. Actin stress fibres, which are anchored to the numerous focal adhesion plaques associated with the invasome structure, are typically found to be twisted around its basal part. The formation of invasomes was found to be inhibited by cytochalasin D but virtually unaffected by nocodazole, colchicine or taxol, indicating that invasome-mediated invasion is an actin-dependent and microtubuli-independent process. Bacterial internalisation via the invasome was consistently observed with several clinical isolates of B. henselae, while a spontaneous mutant obtained from one of these isolates was impaired in invasome-mediated invasion. Instead, this mutant showed increased uptake of bacteria into perinuclear localising phagosomes, suggesting that invasome-formation may interfere with this alternative mechanism of bacterial internalisation. Internalisation via the invasome represents a novel paradigm for the invasion of bacteria into host cells which may serve as a cellular colonisation mechanism in vivo, e.g. on proliferating and migrating endothelial cells during Bartonella-induced vaso-proliferative tumour growth.

Abstract:
The transdermal route is an important pathway for localized or systemic effects. The stratum corneum, the outer layer of the skin, is an essential skin permeation barrier for many drugs. To overcome this barrier, several techniques have been developed, including the use of the vehicle and nanocarriers to improve drug penetration. Recently, different types of nanocarriers have been designed to improve the dermal and transdermal delivery of medicines like ‘INVASOME’. We made clotrimazole loaded invasome gel which is used in fungal treatment. The procedure used in formulation of invasomal gel is mechanical dispersion method. This preparation is evaluate by many parameters they are appearance, spreadability, solubility, in vitro drug diffusion. And also discuss application of invasome.

Abstract:
Although it has limited solubility and poor permeability to the skin, voriconazole (VRC) is a viable choice for the topical treatment of fungal infections. Owing to these constraints, treating the inflection requires several injections over an extended period. The goal of this work was to produce a VRC invasome gel with improved topical antifungal activity. The Box-Behnken design (BBD) program was utilized to optimize the IVS after it was created using the thin-film hydration process. The optimized invasome formulation through BBD has 159.9 nm of vesicle size, 68.58% of entrapment efficiency, and 23.5 mV of zeta potential. The spherical shape of the vesicle was revealed using scanning microscopy. According to FTIR spectra, the medication and polymers do not interact. The optimized invasomal VRC gel exhibited an optimum of 345 to 589 cp. Ex-vivo permeation studies of IG4 exhibited a higher flux of 0.168 as compared to pure voriconazole. An antimicrobial study was accepted to check the antimicrobial efficiency of voriconazole gel (IG4). The study confirmed a good zone of inhibition of the fungus infection (C. albicans). The voriconazole invasome’s potential diffusion and antibacterial efficacy are demonstrated here. Invasomal gel was found to be an effective carrier and a desirable strategy for improving the topical distribution of VRC to treat fungal infections.

Abstract:
Background/Objectives: Raloxifene (RLF) is a therapeutic option for invasive breast cancer because it blocks estrogen receptors selectively. Low solubility, limited targeting, first-pass action, and poor absorption are some of the challenges that make RLF in oral form less effective. This study aimed to create an intra-tumoral in situ pH-responsive formulation of RLF–invasome (IPHRLI) for breast cancer treatment, with the goals of sustaining RLF release, minimizing adverse effects, and enhancing solubility, bioavailability, targeting, and effectiveness. Methods: Numerous RLF–invasome formulations were optimized using design expert software (version 12.0.6.0, StatEase Inc., Minneapolis, MN, USA). Integrating an optimal formulation with an amalgam of chitosan and glyceryl monooleate resulted in the IPHRLI formulation. In vivo testing of the IPHRLI formulation was conducted utilizing the Ehrlich cancer model. Results: Requirements for an optimum RLF–invasome formulation were met by a mixture of phospholipids (2.46%), ethanol (2.84%), and cineole (0.5%). The IPHRLI formulation substantially sustained its release by 75.41% after 8 h relative to free RLF. The bioavailability of intra-tumoral IPHRLI was substantially raised by 4.07-fold compared to oral free RLF. Histopathological and tumor volume analyses of intra-tumoral IPHRLI confirmed its efficacy and targeting effect. Conclusions: the intra-tumoral administration of the IPHRLI formulation may provide a potential strategy for breast cancer management.

Abstract:
Carvacrol is a monoterpenoid phenol found in many essential oils that has antibacterial, antifungal and antiparasitic activities. Drug loaded-invasome systems are used to deliver drugs utilizing nanoparticles to improve bioavailability, efficacy, and drug release duration. As a result, the present study developed carvacrol-loaded invasomes and evaluated their acaricidal effect against Rhipicephalus annulatus (cattle tick) and Rhipicephalus sanguineus (dog tick). Carvacrol loaded-invasome (CLI) was prepared and characterized using UV/Vis spectrophotometer, zeta potential measurements, Scanning Transmission Electron Microscopy (STEM), Fourier Transform Infrared (FT-IR) Spectroscopy, and Differential Scanning Calorimetry Analysis. CLI (5%) induced significant mortality (100%) in R. annulatus adult ticks with LC50 of 2.60%, whereas the LC50 of pure carvacrol was 4.30%. Carvacrol and CLI were shown to have a significant larvicidal action on both tick species, with LC50s of 0.24 and 0.21% against R. annulatus and 0.27 and 0.23% against R. sanguineus, respectively. Carvacrol and CLI (5%) induced significant repellent activities for 24 h against R. annulatus and R. sanguineus, as evidenced by the rod method and the petri-dish selective area choice method, respectively. High-performance liquid chromatography (HPLC) demonstrated that the CLI form had 3.86 times the permeability of pure carvacrol. Moreover, carvacrol and CLI inhibited acetylcholinesterase activity and decreased glutathione and malonedealdehyde levels in the treated ticks. In conclusion, invasomes significantly improved adulticidal and repellency activities of carvacrol against both tick species.

Abstract:
Invasomes are novel vesicular systems that exhibit improved transdermal penetration compared to conventional liposomes. These vesicles contain phospholipids, ethanol, and terpene in their structures; these components confer suitable transdermal penetration properties to the soft vesicles. The main advantages of these nanovesicles lie in their ability to increase the permeability of the drug into the skin and decrease absorption into the systemic circulation, thus, limiting the activity of various drugs within the skin layer. In this paper, several features of invasomes, including their structure, mechanism of penetration, applications, characterization, and potential advantages in dermal drug delivery, are highlighted. Overall, this review suggests that enhanced transdermal penetration of drugs using invasomes provides an appropriate opportunity for the development of lipid vesicular carriers.

Abstract:
Management of fungal infections, for those affecting skin, has faced challenges in poor penetration and efficacy. Traditional topical antifungal therapies often show limited results due to the skin’s barrier properties. Invasomes, has emerged as a greater tactic to improve drug penetration through skin, improving penetration and effectiveness of antifungal drugs. This review comprehensively analyzes invasomes as a transdermal drug delivery system specifically for antifungal therapies. It covers structure, composition, and mechanism of action of invasomes, Focusing on their advantages over conformist drug delivery systems. Key factors influencing invasome efficacy, such as the incorporation of phospholipids, ethanol, and terpenes, are discussed in detail. Invasomes significantly improve drug permeation through skin due to their flexible lipid bilayer structure and presence of ethanol and terpenes, which disrupt skin’s lipid matrix. Various studies have demonstrated the potential of invasomes in delivering antifungal drugs more effectively than conventional topical formulations. The increased drug bioavailability and improved therapeutic outcomes underscore the utility of invasomes. Invasomes characterize a capable transdermal delivery system for antifungal drugs, offering improved drug permeation, stability, and therapeutic efficacy. Their ability to overcome the skin barrier makes them an alternative to traditional topical formulations.

Transdermal drug delivery refers to the drug administration route through the skin that achieves the local or systemic treatment approved for clinical use. Invasomes are novel vesicular systems that exhibit improved transdermal penetration compared to conventional liposomes. These vesicles contain phospholipids, ethanol, and terpene in their structures; these components confer suitable transdermal penetration properties to the soft vesicles. Ethanol, terpenes and phospholipids are the key structural composition, systematize physicochemical properties of invasomes. Nanovesicles can enhance the skin's permeability, which means they allow larger or less penetrative drugs to reach deeper layers of the skin. nanovesicles reduce the drug’s absorption into the bloodstream, helping to keep its effects localized. These vesicular systems, deliver the drugs at predetermined rate by controlling and sustaining release as per the requirement. Furthermore, invaomes used in the treatment of eczema, hypertension, acne, cancer, eosinophilic pustular folliculitis, erectile dysfunction, fungal infections, photodynamic therapy have also been discussed.

Abstract:
Skin cancer is the most frequent cancer throughout the world. Vismodegib (VSD) is a hedgehog blocker approved for the prevention and treatment of skin cancer. VSD, however, is poorly bioavailable and has been linked to side effects. This work focused on designing a nano-invasome gel as a vehicle for enhancing the permeation, bioavailability, and efficacy of VSD. Additionally, the combined effect of terpenes and ethanol was studied on the permeation of VSD compared with liposomes. The prepared VSD-loaded invasomes (VLI) formulation included cineole (1%v/v), cholesterol (0.15%w/w), phospholipid (2%w/w), and ethanol (3%v/v) and displayed an entrapment efficiency of 87.73 ± 3.82%, a vesicle size of 188.27 ± 3.25 nm, and a steady-state flux of 9.83 ± 0.11 µg/cm2/h. The VLI formulation was vigorously stirred into a carbopol base before being characterized in vivo to investigate the permeation, bioavailability, and efficacy of VSD. The VLI gel enhanced the dermal permeation of VSD and, as a result, had 3.59 times higher bioavailability with excellent antitumor action as compared to oral VSD. In summary, as an alternative to oral administration for skin cancer treatment, invasomes are efficient carriers for delivering VSD and enhancing its transdermal flux into deep skin layers.

Abstract:
Invasive Alien Species follow three phases: introduction, establishment and expansion. Few of the introduced species, reach the expansion phase successfully. Many traits are associated to invasiveness of a species and explain their invasion success. These traits vary between species and spatial scales. Despite being one of the most threatened groups, few studies have examined the factors that favour the invasion of amphibian species and its potential impact on native communities. The objective of this thesis was the analysis of the invasiveness of the painted frog (Discoglossus pictus auritus) at macro-, meso- and micro-scale. The species was introduced at final of XIX century in Banyuls sur Mer (France) and currently is expanding both in France and in Spain with abundant populations. Previous studies indicate its potential negative effect on the native community and the importance of increase the knowledge about the painted frog as an invasive species. While macro-scale provide useful predictive information of invasion success, meso-scale defines how species insert in new communities organization and micro-scale which particular traits favor its success. Our results at macro-scale showed that D. pictus introduction accomplished niche conservatism during its introduction. The existence of numerous suitable areas to colonize and the good dispersal abilities to expand throughout more unsuitable areas, predicts a range expansion in the future, mostly to the north because of climatic change. In the invaded area the species occupies mainly ephemeral ponds where presumably would obtain high reproductive success. D. pictus present good abilities to overcome desiccation events, but its plasticity is restricted to resources availability. It also displays phenotypic plasticity facing new predators but milder than the observed in native species. This reduced response could limit reproductive success of D. pictus in ponds with larger duration where predation and competition pressure increase. By last, the painted frog showed good trophic traits that are favourable for invasion of ephemeral ponds and could limit the fitness of a native species (B. calamita) when resources are scarced or there is a risk of pond desiccation. The integrative view derived of the three scales study let us to a better understanding of species invasion.

Abstract:
Invasive species, together with other perturbations like nutrient enrichment, threaten native biodiversity worldwide. This thesis is framed within the context of the invasion of a small fish, the eastern mosquitofish Gambusia holbrooki (Girard, 1859) in ecosystems subject to environmental degradation due to excessive fertilisation with nitrates. By means of experimental and field observational studies, the hypothesis of whether eutrophication mediates the invasive success of G. holbrooki is explored. Specifically, the initial aim is to understand the ecological mechanisms exerted by fish introduction and nitrate pollution in altering the native communities, the ecological processes and their interaction. Nutrient enrichment causing eutrophication, but also physical habitat quality, abundance of predators and parasites prevalence could influence the life history and the invasive success of G. holbrooki. Should the invader fitness be mediated by any factor, managers could carry out an ecosystem approach by favouring such desired conditions against the invader success. Either by direct elimination or through an ecosystemic approach, the loss of G. holbrooki populations should be substituted by other native species, such as Aphanius iberus and provide a similar mosquito control efficiency in clean and eutrophic waters. Results indicate a negligible interaction between the impacts exerted by both fish introduction and nitrate pollution on the recipient ecosystem. The loss of fish biomass in high nitrate concentration would be attributed to indirect effects such as a decline in prey abundance, rather than a direct alteration on fish physiology. This low susceptibility of G. holbrooki to nitrates, especially in females, confirms its high tolerance to a wide gradient of environmental conditions reported by previous authors. Moreover, the low prevalence of parasites and the high variability of life history traits, mainly attributed to G. holbrooki density in the pond over other environmental factors leave managers with low chances of controlling the invader apart from its eradication. Given the similar but slower efficiency of the native community both in eutrophic and clean waters in reducing mosquito abundances and the difficulty of eradicating G. holbrooki once established, favouring the presence of native competitors and preventing further introductions of G. holbrooki for biological control seems a priority.

The use of riding animals in natural areas has caused great concern because their use cause environmental impacts.
The aim of this thesis is to provide the first data on the ecology of European Catfish (Silurus glanis) introduced to the Iberian Peninsula and to appraise its ecological impact on native biota. We sampled the fish assemblage of 14 Catalan reservoirs (Spain). The European catfish is nowadays introduced in four Iberian river basins: in the Ebro river basin for 30 years, in the Ter and Tajo river basin for a few years and we report the first record for the Llobregat river basin. We demonstrate the utility of comparing early and late invasion stages to understand the ecological changes caused by invasive species: recent introductions of catfish had smaller and younger catfish with growth rates higher than old and native populations and preying on fish instead of crayfish for old introductions. We also report the first published telemetry data for catfish.

Abstract:
The introduction of species in places outside their natural distribution is an important concern in biodiversity conservation. Callithrix aurita is endemic in regions of high-altitude forests of the Atlantic forest of southeastern Brazil. The most relevant criteria of fitting as endangered species are: habitat destruction, inability to adapt to degraded secondary forests, population decline, restricted distribution and introduction of invasive alien species. These criteria, coupled with the apparent rarity, explain its inclusion on the Official List of Species of Brazilian Fauna Threatened with Extinction. The objectives are: to estimate the population size of C. aurita, C. penicillata and their hybrids in the Serra dos Órgãos National Park, assess hybridization between species by morphology and laboratory check the health status and confirm the involvement of C. aurita in the fatherhood of trapped animals, propose a plan for eradication and control of invasion of C. penicillata in the Park. The population sizes of the two primate species were estimated by the method "Distance Sampling". A total of seven marmosets were captured with live traps set for the chemical and physical restraint and subsequent completion of the procedures. For the haemogram, the biochemical and genetic analysis, blood was collected in a test tube containing anticoagulant and kept at refrigerator temperature until the moment of handling / processing of samples. Callithrix aurita appears to be well preserved only in the park area corresponding to the portion located in the city of Petrópolis. The cytogenetic and molecular analysis of hybrids are a useful tool to confirm whether or not hybridization, identifying the species involved and seeing if there is a tendency in the backcrosses. One may suggest that there is a tendency for species differentiation and identification of hybrid individuals by standard hematological and biochemical, to be confirmed with a larger sample of the species C. aurita, preferably from the same locality and under the same conditions. In the case of C. aurita, the main recommendations for its conservation research to include the registration of other populations in areas of distribution free of intrusion, so you can evaluate the chances of population recovery and species survival. The creation of new protected areas should be encouraged, as well as more detailed studies on the species already known sites of occurrence, and a safe program of captive breeding.

Abstract:
Compararam-se, ainda, os micro-habitats usados por ambas as espécies num meio urbano com grande densidade e influência humana; Abstract: First assessment on the introduction of the Italian wall lizard (Podarcis siculus) in Parque das Nações (Lisbon) Currently, animal biological invasions are one of the main threats to biodiversity worldwide and, consequently, one of the biggest problems in nature conservation. Known the introduction of the Italian wall lizard Podarcis siculus in the area of Parque das Nações, Lisbon, were evaluated both the current territorial extent of the introduced individuals and the degree of spatial segregation exerted by the invasive population upon the native species, Podarcis virescens. The obtained results suggest a scenario of non-expansion of the Italian wall lizard beyond the area of known occurrence. However, this interferes with the local distribution pattern of P. virescens. The microhabitats used by both species were also compared in an urban environment with great human density and influence.

Abstract:
Invasive species have advantageous traits over native species which may turn them into one of the most important threats to biodiversity. Wild boar (Sus scofa) is considered one of the worst invasive species of the world due to the high impact over natural ecosystems. Worldwide, hunting is the most used control strategy with variable efficacy. In order to determine the efficacy of a control strategy it is necessary to evaluate the invasive population traits and the hunting impact. This work aims to studying the legal hunting effectiveness as a control method of wild boar population at the National Forest of Capão Bonito, São Paulo, Brazil. Cameras traps were used to estimate group size and transects were used to detect vestiges in the study area. Population characteristics were obtained from hunted specimens and from literature review. The hunting pressure was informed by the hunter and neighbors provided additional information about damages caused by the species. There is evidence of the wild boar population reduction after the beginning of hunting. However, there are no sufficient quantitative evidence about such impact. The accuracy of the results presented in this work could be improved by the development of further research in population and behavioral characteristics including the potential impact of illegal hunting.

Abstract:
Research suggests that the threat from invasive non-native species (INNS) is growing. Biological invasions by INNS harm native species and habitats and can have economic impacts. Biosecurity measures can be adopted to prevent the introduction and spread of INNS. This POSTnote summarises the drivers and impacts of INNS and the measures needed to meet national and international environmental targets.
