Background: Aim of this study was to evaluate the efficacy of PCR targeting IS1081in diagnosis of pediatric tuberculosis and compare the results with MGIT culture.Methods: This prospective study was conducted in the department of pediatrics, S.N. medical college, Agra. 100 subjects (28 pulmonary 72 extra pulmonary) were registered in study. The specimens obtained from these cases were subjected to Ziehl–Neelsen staining (ZN), MGIT 960 TB culture and PCR targeting insertion sequence IS1081. Sensitivity, specificity, PPV and NPV of PCR were calculated in pulmonary and extra pulmonary specimens.

ABSTRACT The two haloacetate dehalogenase genes, dehH1 and dehH2, on the 65-kb plasmid pUO1 from Delftia acidovorans strain B were found to be located on transposable elements. Abstract:

A PCR test based on insertion sequence IS1081 was developed to detect Mycobacterium tuberculosis complex organisms in the peripheral blood. The method was applied to blood samples from immunocompetent individuals with localized pulmonary tuberculosis. Seven of 16 (43.75%) blood samples were found to be positive for the circulating DNA copies of M. tuberculosis complex.

It has been observed that the class III bacteriophages Mu and D108 and the class II elements, Tn3, Tn1000, IS101 and Tn951, share the characteristics of replicatively transposing to random positions, producing 5 bp target site duplications and of containing the sequence 5$ sp prime$-PUCGAAAPu-3$ sp prime$ at bp 21 from their ends. These shared characteristics led to the hypothesis that these class III and class II elements evolved from a common ancestor transposon. Iss1rs est responsable de la co-integration entre le pucl22 et le plasmide conjugatif pva797. Apres resolution des co-integrats pucl22::pva797, chaque pva797 conserve une iss1rs. Iss1rs s'insere dans une cible de 8 bp du pva797 et a 808 pb de long. Elle possede des extremites inversees de 18 pb. L'orf1 d'iss1rs correspond a une transposase de 226 acides amines.
