This thesis presents research into the development of a novel conjugate-addition/cyclisation methodology for the synthesis of isoindole derived nitrones. To begin, a review of the principle methods for synthesising nitrones is reported, followed by a discussion of the current uses of nitrones and the requirement for straightforward syntheses of nitrones contained within an isoindole scaffold. The first section of this work describes the synthesis of a library of novel rhein analogues that are potential anticancer agents. The design of these compounds takes advantage of the ability for rhein to intercalate into DNA and as the incorporation of an alkylating agent, which serves to covalently modify DNA. Several carbon-carbon bond forming methodologies, including the first examples of Heck and Sonogashira coupling applied to the isoindoline nitroxide class, were utilised to give novel robust aromatic frameworks. Palladium-catalysed Heck coupling of brominated nitroxides and ester-substituted olefins generates novel nitroxides possessing extended conjugation.
