Healthy EBV seropositive donors tested for the ability to mount Th responses against LMP1 responded with secretion of high levels of the immunosuppressive cytokine IL-10 which was secreted from cells phenotypically analogous to the Tr1 class of regulatory T-cells.  The epitopes inducing this IL-10 secretion were clustered in the hydrophobic, transmembrane half of the protein that corresponded to a cluster of high affinity MHC class II binding domains.  Since the LMP1 induced TO cells could effectively suppress immune responses in an IL- 10 dependent manner, it seems likely that the induction o

A targeting peptide at the N-terminus of a precursor protein usually directs the protein synthesized in the cytosol to a specific organelle in the cell. Interestingly, some targeting peptides, so-called dual targeting peptides (dTPs) can target their protein to both mitochondria and chloroplasts. In order to understand the mechanism of dual targeting, a dTP from threonyl tRNA synthetase (ThrRS-dTP) was investigated as a model dTP in this thesis work. The results suggest that ThrRS-dTP is intrinsically disordered in solution but has an α-helical propensity at the N-terminal part. Tom20 and Toc3

Abstract:
Recently, linear ubiquitin assembly complex (LUBAC)-mediated linear ubiquitination has come into focus due to its emerging role in activation of NF-κB in different biological contexts. However, the role of LUBAC in LMP1 signaling leading to NF-κB and interferon regulatory factor 7 (IRF7) activation has not been investigated. We show here that RNF31, the key component of LUBAC, interacts with LMP1 and IRF7 in Epstein-Barr virus (EBV)-transformed cells and that LUBAC stimulates linear ubiquitination of NEMO and IRF7. Consequently, LUBAC is required for LMP1 signaling to full activation of NF-κB

Epstein-Barr virus (EBV)-encoded latent membrane protein 1 (LMP1) plays important roles in EBV-mediated B cell transformation, development of EBV-associated malignancies, and exacerbation of certain autoimmune conditions. LMP1 functionally mimics tumor necrosis factor receptor (TNFR) superfamily member CD40, but LMP1 signals are amplified and sustained compared to those induced by CD40. CD40 and LMP1 rely on TNFR-associated factors (TRAFs) to mediate signaling, but use TRAFs differently. TRAF6 is important for CD40 signaling, and was implicated in LMP1 signaling in non-B cells.
