Abstract:
The fact that optical force is very significant in the microscopic world and can be used to manipulate microparticles has triggered an evolution in micromanipulation, in particular, the manipulation of biological species and colloidal particles. The induced optical force can easily be more than 103 times of the particle's weight. The particle size that are accessible to optical forces ranges from tens of nanometers to hundreds of micrometers. One of the most well-known tools in optical manipulation is called optical tweezers, which is, in essence, performing optical trapping by a strongly focu

Inhalation of respirable coal mine dust (RCMD) can lead to chronic lung diseases, including coal worker’s pneumoconiosis (CWP) and more severe forms such as progressive massive fibrosis. After the Federal Coal Mine Health and Safety Act was passed in 1969, limits on exposure to respirable dust were set, and the prevalence of CWP abruptly decreased. However, during the last two decades, a resurgence of the disease has been reported. Light-controlled manipulation of neuronal activity has transformed the field of neurobiology. Light provides temporal and spatial resolution over activation or inhibition of targeted populations of neurons, one single neuron or single synapses. Such precision could be achieved with optogenetics, which is based on the over-expression of light-sensitive proteins, but it requires gene therapy and often alters cellular physiology. An alternative to optogenetics is offered by photopharmacology (the development of light-regulated drugs or photoswitches), which can operate on endogenous receptors wit

Imaging at the nanoscale and/or at remote locations holds great promise for studies in fields as disparate as the life sciences and materials sciences. One such microscopy technique, stimulated emission depletion (STED) microscopy, is one of several fluorescence based imaging techniques that offers resolution beyond the diffraction-limit. All current implementations of STED microscopy, however, involve the use of free-space beam shaping devices to achieve the Gaussian- and donut-shaped Orbital Angular Momentum (OAM) carrying beams at the desired colors –-- a challenging prospect from the stand

In this thesis, studies on the applications of digital micro-mirror devices (DMD) to enhancement of digital optical microscope images are presented. This involves adaptation of the fast switching capability and high optical efficiency of DMD to control the spatial illumination of the specimen. The first study focuses on a method of using DMD to enhance the dynamic range of a digital optical microscope. Our adaptive feedback illumination control method generates a high dynamic range image through an algorithm that combines the DMD-to-camera pixel geometrical mapping and a feedback operation.
