Abstract:

Abstract:
The mammary gland of lactating transgenic animals has several advantages for production of heterologous proteins including a high cell density that results in high concentrations of secreted protein and the ability to perform several types of post-translational modifications. Transgenes were constructed from the 4.1 kbp murine Whey Acidic Protein promoter (mWAP) and the three cDNAs coding for the Aα, Bβ and γ fibrinogen chains to evaluate the requirements of the transgenic murine mammary gland for high level secretion of fully assembled human fibrinogen. Four studies were conducted to investigate the effect and site of action of exogenous and endogenous opioids on pituitary growth hormone (GH) and prolactin (PRL) secretion in Holstein calves. In the first study, the effect of the opioid agonist DAMME (D-Ala²⁺,N-Me-Phe⁴,Met(O)⁵-01 enkephalin) on plasma GH and PRL secretion was measured in Holstein calves in fall season. Plasma concentrations of both GH and PRL increased in response to DAM ME injection. The mammary alveolar cell-T (MAC-T) cell line is able to uniformly differentiate and secrete casein proteins in response to dexamethasone, insulin and prolactin and is extensively used to study bovine mammary epithelial cell function. Growth hormone (GH) has been shown to increase milk protein synthesis both in vivo and in mammary cell models, and induce cytoskeletal rearrangement in 3T3 fibroblast cell line and a Chinese hamster ovary (CHO) cell line. Few studies have focused on identifying the mechanisms involved in differentiated MAC-T cells’ response to GH. We tested the hypothesis that MA

The ability of mammary tissue from prepubertal bulls and heifers of beef and dairy breeds to respond to hormonal stimuli through synthesis and secretion of milk proteins was studied. Mammary tissue was explanted and cultured for 96 h in basal medium (B) which contained hormones necessary for maintenance, or stimulatory medium (P), further supplemented with prolactin. However, the major limitation of the microorganisms is possibly lack of post-translation modification that may affect the stability and activity of produced proteins.
