Brassica napus microspores can be diverted from pollen development toward haploid embryo formation in culture by subjecting them to a heat stress treatment. This switch in developmental pathways has been shown to be accompanied by the induction of high levels of napin seed storage protein gene expression (DeMoor, 1992). Specific members of the napin multigene family that were expressed at this time were cloned from a cDNA library prepared from microspores that had been induced to undergo embryogenesis. Dissolution of the pollen tetrad walls after meiosis, and subsequent release of the haploid microspores, is essential for pollen development in many species, including Arabidopsis thaliana. Despite the importance of microspore release, not all the wall-degrading enzymes involved have been identified in A. thaliana. 18 endo-β-1,3-glucanase genes were identified in A. thaliana that are expressed in buds at microspore release. 碩士
國立中興大學
農藝學系所
97
Purple coneflower (Echinacea purpurea L.) is one of the most popular health protection herbs recently and it is a cross-pollinated crop with dominant self-incompatibility. Considering the heavy burden of purification of inbred line via self-pollination in a long schedule, we tried to employ anther culture to get doubled-haploid plants. To promote efficiency the microspore culture was studied for avoiding paying attention to dissect where the regenerated plants were derived from. Furthermore, karyotype examination protocol was developed to aim at coneflower plantl

Abstract:
- Ph.D. thesis - Milan Urban, 2017 In some years, the agricultural production of oilseed rape, an important crop in the Czech Republic, is - besides biotic stress - facing the problem of damage caused by frost or drought. Together with special attention paid to proteins revealing responses between crop genotypes with differential abiotic stress tolerance levels we reviewed possible applications of proteomic results in crop breeding programs aimed at an improvement of crop stress tolerance (paper 1). For first original result, cold temperature was imposed upon non-vernalized plants in the stage
