Abstract:
The aims of this study were to develop polymeric NP formulations for triamcinolone acetonide (TA) delivery, from biodegradable and biocompatible hydrophobic polymers, which provide sustained release, prolonged stability and low toxicity, and to assess the toxicity of TA NPs (TA-NPs) compared to TA alone upon BALB/c 3T3 and ARPE 19 cell culture models. The study involved investigation of three different types of polymers: poly(D,L,lactide) (PDLLA), poly(D,L,lactide-co-glycolide)(PLGA) and methoxypolyethyleneglycol poly(D,L,lactide-co-glycolide)(mPEG PLGA). Self- or driven-assembly of nanoparticles (NPs) into mesoscopic ordered structures play a crucial role in a large variety of applications including energy, pharmaceutical, food, drug delivery, immunology and technological. On the one hand, trying to prevent and avoid the self-organization of nanoparticles has traditionally been the main issue to stabilizing nanosuspensions, foams, and emulsions. On the other hand, the aggregation of building-blocks into mesoscopic structures has allowed to explore new materials with desired functionalities and properties. Abstract:
The properties of polymer–nanoparticle (NP) mixtures significantly depend on the dispersion of the NPs. Using molecular dynamics simulations, we demonstrate that, in the presence of polymer–NP attraction, the dispersion of NPs in semidilute and concentrated polymers can be stabilized by increasing the polymer concentration. A lower polymer concentration facilitates the aggregation of NPs bridged by polymer chains, as well as a further increase of the polymer–NP attraction. Evaluating the binding of NPs through shared polymer segments in an adsorption blob, we derive a linear relationship betwe

Protein replacement therapy is to-date one of the main clinical approaches to treat a wide number of diseases that are correlated to a protein dysfunction or absence. However, the systemic administration of proteins rarely translates into a successful therapy, due to the fragile nature of these molecules and their inability to reach specific targets in vivo. Pharmaceutical technology strategies such as the encapsulation into a nanovector could overcome the limits of protein administration, since nanovectors can protect the payload from degradation and rapid clearance, and they can be f
