Obligate intracellular pathogens must acquire host cell-derived nutrients to facilitate their survival. One such bacterial pathogen, Anaplasma phagocytophilum, replicates within neutrophils and non-phagocytic cells in a bacterial-modified, host cell-derived vacuole. The bacterium exploits host cell vesicular trafficking pathways to route nutrients to its vacuole and utilizes Rab GTPases, guanine nucleotide-dependent, vesicular trafficking regulators, to do so. Rheumatoid cachexia (RC), i.e. muscle wasting and adiposity gain in rheumatoid arthritis (RA), is driven by inflammation and is a major contributor to reduced strength and physical function. Current treatment for RA is based on approaches that aim to tightly control inflammation from diagnosis onwards. The influence of zinc status on the expression of p53, the human tumor suppressor gene as well as downstream target genes of p53 were examined in human cell lines. HepG2 cells were depleted of cellular zinc using a low-zinc media containing Chelex-100 treated serum. Cellular zinc levels were depleted to 64% of controls (4.0 μM zinc, comparable to normal media). p53 mRNA was increased in the zinc-depleted HepG2s as compared to control, however, p53 protein levels and DNA binding activity were not significantly different among treatment groups. We determined whether children older than 1 year from non-western immigrant families had lower serum 25-hydroxyvitamin D levels than children from western born families. Children ages 1-6 years were recruited through the TARGet Kids! practice based research network. Univariable analysis revealed that non-western immigrant children had 4 nmol/L lower 25-hydroxyvitamin D (p=0.006; 95% CI:1.4-8.0) and increased odds of 25-hydroxyvitamin D <50 nmol/L (OR 1.9, 95% CI:1.3–2.9).
