L'epissage des arn premessagers chez la levure se fait au sein d'un large complexe ribonucleoproteique dont l'assemblage necessite l'intervention de nombreuses proteines. L'une d'entre elles, prp4, est retrouvee au niveau des snrnps u4/u6 et u4/u6-u5, mais aussi dans le spliceosome avant la premiere etape de la catalyse. Elle quitte le spliceosome juste avant la premiere etape d'epissage, au moment ou le snrna u4 se detache du complexe. Phenotypic studies of mice with altered levels of PrPc expression showed differences in lymphocyte subset composition of peripheral lymphoid tissue. Mice with a complete absence of PrPc expression showed a reduction in mononuclear cell numbers in lymphoid tissue compared to wild type mice. FACS analysis of these cells from mice that expressed different levels of PrPc revealed no difference in subset composition. Cellular prion protein (PrPsen ou PrPc) is expressed in a wide variety of cells, specialy neurons. This protein is rich in alpha helix structures, but it can assume an alternative conformational isoform known as PrPres (or PrPsc). PrPres presents mainly beta sheet structure and gains the ability to form aggregates as well as is protease resistant. PrPres is able to propagate by converting PrPsen molecules to the alternative conformation isoform PrPres in a new biologic mechanism, not yet fully understood. Four dominant negative mutants of PRP2 were produced by site-directed mutagenesis of the conserved ATP-binding motif (Motif A) of PRP2. These mutants, when overproduced in a wild-type yeast strain caused a growth inhibition due to inhibition of splicing, leading to an accumulation of pre-mRNA. <I>In vitro</I> analysis of cell-free splicing extracts showed that splicing was inhibited in the presence of the dominant negative proteins. The Prp2 protein of <I>Saccharomyces cerevisiae </I>is a 100kDa non-snRNP splicing factor with amino acids common to the DEAD/H-box family of ATP-dependant RNA helicases. Prp2, Prp16 and Prp22 proteins have extensive sequence similarity, and function at sequential steps in the splicing pathway. Prp2 and Prp16 form transient reactions with the spliceosome, Prp2 at step 1 and Prp16 at step 2.
