This thesis comprises of two different studies investigating rotavims infections among children conducted in two Saudi cities namely Maddina and Oizan. Enzyme immunoassays were used to examine faecal samples obtained from children with acute, dehydrating gastroenteritis in the presence of rotavirus on both studies. In addition, all Maddina samples were subjected to Electron Microscopy (E.M) to detect any non-group A rotavims and all other viral enteropathogens. The rotaviral genome was then extracted using a guanidine isothiocyanate method. Rotavimses were then examined by polyacrylamide gel e

Background: Rotavirus is one of the most common diarrheal diseases in children less than 5 years of age. Rotavirus vaccines have greatly reduced this burden in the United States. An examination was conducted to determine possible disparities in RV vaccination rates compared to DTaP. Methods: Children were actively enrolled during two rotavirus seasons from January-June of 2010 and 2011 in the Emergency Departments (ED) and inpatient floors from all Children's Healthcare of Atlanta (CHOA) sites (Scottish Rite, Egleston, and Hughes Spalding) with acute gastroenteritis (AGE). Abstract:
Hajj is the annual gathering of Islam practitioners in Mecca, Saudi Arabia. During the event, gastrointestinal infections are usually experienced and outbreaks have always been a concern; nevertheless, a deep and integrative study of the etiological agents has never been carried out. Here, I describe for the first time the epidemiology of pathogenic enteric viruses during Hajj 2011, 2012 and 2013. The focus of this study was the common enteric viruses Astrovirus, Norovirus, Rotavirus and Adenovirus. An enzyme Immunoassay established their presence in 14.9%, 15.0% and 6.6% of the reported cases

Abstract:
Group A rotaviruses are a major cause of acute gastroenteritis in children. Almost all children are infected by the age of 5 years old. Rotavirus disease causes around 600,000 deaths per year. VP4 (P) and VP7 (G) genotypes were analyzed for prevalence and potential antigenicity, as they are known to elicit a neutralizing antibody response during infection. This study predicted the effectiveness of two recently licensed rotavirus vaccines based on Canadian surveillance. 271 out of 348 diarrhea samples from 8 paediatric hospital were successfully genotyped by PCR.
