La maladie de Parkinson est une pathologie neurodégénérative qui peut être liée à la présence de mutations génétiques au niveau du gène SNCA codant pour le pro oxydant α-synucléine. De nombreux travaux montrent que la levure Saccharomyces cerevisiae constitue un modèle utile et valable pour étudier les mécanismes moléculaires de la toxicité cellulaire de l'α-synucléine. Abstract:
This thesis investigated the role of genetics in schizophrenia. Particular schizophrenia risk genes were examined to determine their role in the development of specific schizophrenia symptoms. These genes were further studied to determine how they interact to cause schizophrenia symptoms. One important schizophrenia risk gene (BDNF) was studied in detail to determine its effect on gene expression and epigenetic changes in brains of schizophrenia individuals. This thesis has contributed to a better understanding of the role of genetics in the development of particular symptoms of schizophrenia.

Cyclic dipeptides are widely used as models for larger peptides because of their simplicity and limited conformational freedom. Some cyclic dipeptides have been shown to produce antiviral, antibiotic and anti-tumour activity (Milne et al., 1998). Nesta dissertação o processo de adsorção da tiossemicarbazona do formilferroceno (TFF) em eletrodos de prata e ouro, em soluções aquosas 0,1 mol.L-1 de KCl e de acetonitrila. 0,1 mol.L-1 de NaClO4, foi caracterizado através da técnica espectroscópica SERS. Verificou-se através das variações espectrais que a adsorção da TFF ocorre através dos átomos N1 do grupo imínico e do S do grupo tiocarbonílico. This thesis focuses on understanding implications of nanomaterial quality control and mass transport through internally etched silica coated nanoparticles for direct and quantitative molecular detection using surface enhanced Raman scattering (SERS). Prior to use, bare nanoparticles (partially or uncoated with silica) are removal using column chromatography to improve the quality of these nanomaterials and their SERS reproducibility. Separation of silica coated nanoparticles with two different diameters is achieved using Surfactant-free size exclusion chromatography with modest fractionation.

A bioassay technique, based on surface-enhanced Raman scattering (SERS) tagged gold nanoparticles encapsulated with a biotin functionalised polymer, has been demonstrated through the spectroscopic detection of a streptavidin binding event. A methodical series of steps preceded these results: synthesis of nanoparticles which were found to give a reproducible SERS signal; design and synthesis of polymers with RAFT-functional end groups able to encapsulate the gold nanoparticle. SERS is a useful spectroscopic technique that was discovered 30 years ago, and has recently seen a renaissance in research. Sphere segment void (SSV) substrates have been developed as reproducible, stable SERS substrates by electrodeposition of a metal through a colloidal template. The effect of adsorbing an organic monolayer on the surface of an SSV substrate was studied, which results in a slight shift in the plasmonic absorption.
