Myocytes of varying orientation across the ventricular wall are grouped into layers, separated by cleavage planes. The role of extracellular matrix, in particular collagen, in this laminar organisation has not been fully delineated due to an inability to truly appreciate its 3D relationships. Theoretical studies have demonstrated a pronounced effect of red blood cell (RBC) spacing on tissue oxygen supply. In spite of this, realistic values for RBC spacing and related capillary hematocrit (Hct) are not known in the heart. One of the possible reasons is the lack of proper methodology. Thus, the goal of this research study was twofold: (i) to develop a method to rapidly freeze rat heart in situ, following which RBCs and capillary walls could be simultaneously visualized, and (ii) to apply this methodology in rat hearts to establish whether differences exist in capillary Hct and RBC sp

Taurine is an amino acid found in high concentration (20-30 mM) in mammalian heart. Treatment of rats with the transport antagonist guanidinoethyl sulfonate (GES) depletes cardiac taurine ($>$70%). GES treatment caused a selective prolongation of the QT interval which was correlated with the degree of myocardial taurine depletion (r$ sp2$ = 0.92, p $<$ 0.001). Aging is characterized by a progressive decline in cardiac function. A critical contributor to the age-related impairment in heart function is the loss of cardiac myocytes through ??apoptosis??, or programmed cell death. A dramatic increase in the rate of apoptosis has been reported with aging in the rat left ventricle. In contrast, exercise training not only improves cardiac function, but also reduces the risk of heart disease. However, the ability of exercise training to modulate apoptotic signaling and apoptosis in the aging heart remains unknown. Therefore, the purpose of this study was to

Magnesium (Mg24) is essential for normal functioning of the heart. It is the second most abundant divalent cation in living cells and plays vital roles in cellular regulation. Preservation of intracellular Mg 24 concentration ([Mg24]) is essential in the heart. The concentration of ionized Mg 24 is similar in both the extracellular and intracellular milieu of cardiac myocytes although a large electrochemical gradient exists which will tend to promote Mg24 influx across the sarcolemmal membrane. Therefore homeostatic mechanism(s) must exist which will extrude Mg 24 against this gradient. The ex
