In two separate experiments, weight adjusted doses of nocodazole and acrylamide were injected intraperitoneally at various time intervals into twelve week old female mice. Within the nocodazole experiment, the doses were injected at varying time intervals before and after mating. A new generation of thalidomide survivors have been reported in Brazil, where the drug is used to treat leprosy complications and multiple myeloma. 5-Bromo-2'-deoxyuridine (BrdU), a thymidine analogue, is genotoxic and teratogenic. The exposure of mouse embryos to BrdU at doses that cause malformations induces oxidative stress and an embryonic stress response characterized by an increase in c-Fos dependent AP-1 DNA binding. The goal of this thesis was to test the hypothesis that development is disturbed at sites where BrdU is incorporated into DNA, triggering oxidative stress and c-Fos induction. This investigation evaluated the potential protective role of Zn, through modulation of MT, against the teratogenic effects of isotretinoin (ITR), a dermatological drug which causes oxidative damage, in CD-1 mouse embryos. Significant induction of MT by Zn supplementation was observed in mouse embryos both in vivo and in vitro. On gestational day (GD) 6.5, pregnant mice received a subcutaneous Zn injection of 0 (saline), 20 and 40 mg/kg, resulting in embryonic MT concentrations of 12.5, 54.5 and 93.4 $ mu$g/g protein, respectively, after 48h.
