Doctor of Philosophy Department of Anatomy and Physiology Timothy I. Musch Nitric oxide (NO) is synthesized via distinct NO synthase (NOS) enzymes and constitutes an essential cardiovascular signaling molecule. Whereas important vasomotor contributions of endothelial NOS (eNOS) have been well-described, the specific vasomotor contributions of nNOS-derived NO in healthy subjects during exercise are unknown. The purpose of this dissertation is to test the global hypothesis that nNOS-derived NO is a critical regulator of exercising skeletal muscle vascular control. Specifically, we utilized the selective nNOS inhibitor S-methyl-L-thiocitrulline (SMTC) to investigate the effects of nNOS-derived NO on skeletal muscle vascular function within established rodent models of exercise performance. The first investigation (Chapter 2) identifies that nNOS inhibition with SMTC increases mean arterial pressure (MAP) and reduces rat hindlimb skeletal muscle blood flow at rest whereas there are no effects during low-speed (20 m/min) treadmill running. In Chapter 3 it is reported that nNOS inhibition with SMTC reduces blood flow during high-speed treadmill running (>50 m/min) with the greatest relative effects found in highly glycolytic fast-twitch muscles and muscle parts. Chapter 4 demonstrates that nNOS-derived NO modulates contracting skeletal muscle blood flow (increases), O2 consumption (VO2, increases), and force production (decreases) in the rat spinotrapezius muscle and thus impacts the microvascular O2 delivery-VO2 ratio (which sets the microvascular partial pressure of O2, PO2mv, and represents the pressure head that drives capillary-myocyte O2 diffusion). In Chapter 5 we report that systemic administration of the selective nNOS inhibitor SMTC does not impact lumbar sympathetic nerve discharge. This reveals that the SMTC-induced peripheral vascular effects described herein reflect peripheral nNOS-derived NO signaling as opposed to centrally-derived regulation. In conclusion, nNOS-derived NO exerts exercise-intensity and muscle fiber-type selective peripheral vascular effects during whole-body locomotor exercise. In addition, nNOS-derived NO modulates skeletal muscle contractile and metabolic function and, therefore, impacts the skeletal muscle PO2mv. These data identify novel integrated roles for nNOS-derived NO within healthy skeletal muscle and have important implications for populations associated with reduced NO bioavailability and/or impaired nNOS structure and/or function specifically (e.g., muscular dystrophy, chronic heart failure, advanced age, etc.).

Abstract:

Résumé Le manganèse (Mn2+) est un élément essentiel du corps humain. Ses propriétés paramagnétiques permettent son utilisation comme agent de contraste pour l'IRM (Mn-MRI ou MEMRI). Analogue du calcium (Ca2+), il pénètre les neurones essentiellement par les canaux calciques. Il est ensuite transporté le long des microtubules jusqu'aux synapses où il est libéré, puis capturé par les autres neurones. Ainsi, il peut rendre compte du transport axonal antérograde et rétrograde. L'approche MEMRI peut ainsi apporter des informations uniques sur la connectivité fonctionnelle cérébrale. Toutefois, deux problèmes limitent l'emploi de ce puissant outil d'imagerie in vivo : (i) A doses élevées, le Mn2+ est toxique pour l'organisme et peut provoquer une atteinte grave du système nerveux central, appelé manganisme. Les niveaux et les mécanismes de toxicité sont mal connus. (ii) Le mode de transport du manganèse dans l'approche MEMRI est mal connu. Afin d'apporter des éléments de réponse à ces deux problèmes, nous avons entrepris une étude couplant IRM et microscopie synchrotron pour mieux comprendre le comportement du Mn2+ in vivo. Nous avons précisé les distributions cellulaire et sub-cellulaire du Mn et d'autres métaux pour un modèle de cellules de type neuronal (lignée de neuroblastome N2A), pour des cultures primaires de neurones hippocampiques, mais aussi au niveau de coupes d'hippocampe de rats. En parallèle, nous avons étudié les effets du Mn sur le métabolisme cérébral par une technique de RMN-HRMAS du proton. Pour compléter ce travail, nous avons mis en œuvre l'imagerie MEMRI chez les souris KO MAP6 présentant un déficit d'une protéine stabilisatrice des microtubules pour évaluer la connectivité fonctionnelle du tract thalamo-cortical. Abstract:

Pendant le développement, les projections axonales à longue distance se ramifient pour se connecter à leurs cibles. L’établissement et le remodelage de ces connexions est notamment régulé par l’activité neuronale. L’adaptation de la morphologie de l’axone nécessite alors des quantités importantes de matériel sécrétoire et de facteur trophiques comme le BDNF (brain derived neurotrophic factor). Ce matériel est transporté dans des vésicules le long de l’axone depuis le corps cellulaire où il est synthétisé, vers les sites actifs à l’extrémité de l’axone. Si le relargage de vésicules sécrétoires à la synapse est bien étudié, les mécanismes régulant le transport axonal par l’activité sont encore méconnus.Dans ce travail de thèse, nous avons dans un premier temps développé des outils permettant d’étudier les dynamiques intracellulaires dans des réseaux neuronaux. Nous avons ainsi développé une chambre microfluidique permettant de reconstruire in vitro des réseaux neuronaux physiologiques et compatibles avec de la vidéomicroscopie à haute résolution. Nous avons caractérisé l’établissement et la maturation du réseau et validé l’intérêt de ce dispositif microfluidique dans le contexte de la maladie de Huntington. Nous avons ensuite étudié l’évolution des dynamiques intracellulaires avec la maturation du réseau. Nous avons notamment observé une augmentation du transport axonal de vésicules sécrétoires en fonction de l'état de maturation du réseau neuronal. Ces premières observations ont renforcé l’hypothèse d’une régulation directe du transport axonal de vésicules sécrétoires par l’activité neuronale au cours du développement du réseau.Nous avons ainsi fait évoluer la plateforme microfluidique par l’ajout d’un réseau d’électrodes (MEA) qui permet d'étudier les dynamiques intracellulaires tout en contrôlant l’activité neuronale. A l’aide de ce système, nous avons identifié un groupe de vésicules sécrétoires ancré le long de l’axone et recruté en réponse à une haute activité neuronale en direction des sites présynaptiques actifs. Nous avons alors identifié les acteurs impliqués dans ce mécanisme dépendant de l’activité. Nous avons montré que la myosine Va permettait l’attachement des vésicules le long de l’axone dans des structures d’actine dynamique. L’activité neuronale induit une augmentation de calcium le long de l’axone, via l’activation des canaux calciques dépendant du voltage, qui régule la myosine Va et entraine le recrutement des vésicules stockées dans l’axone sur les microtubules. Une fois les acteurs identifiés, nous avons pu mettre en évidence le rôle de ce mécanisme dépendant de l’activité dans la formation de branches axonales pendant le développement. Enfin, nous avons confirmé l’existence de ce groupe de vésicules dépendant de l’activité et résidant dans l’axone in vivo grâce à la mise au point d'un système d’étude du transport axonal sur tranches aigües de cerveau en microscopie biphotonique.L’ensemble de ce travail propose de nouveaux outils in vitro et in vivo pour comprendre les régulations des dynamiques intracellulaires dans des réseaux neuronaux physiologiques. Grâce à ces outils, nous avons identifié un mécanisme de régulation local qui permet l'adressage rapide de facteurs trophiques vers les branches en développement en réponse à l’activité neuronale During postnatal development, long-distance axonal projections form branches to connect with their targets. Establishment and remodeling of these projections are tightly regulated by neuronal activity and require a large amount of secretory material and trophic factors, such as brain derived neurotrophic factor (BDNF). Axonal transport is responsible for addressing trophic factors packed into vesicles to high demand sites where mechanisms of secretion are well-known. However, mechanisms controlling the preferential targeting of axonal vesicles to active sites in response to neuronal activity are unknown.In this work, we first developed tools to study intracellular dynamics in neuronal networks. We thus developed a microfluidic chamber to reconstruct physiologically-relevant networks in vitro which is compatible with high resolution videomicroscopy. We characterized the formation and maturation of reconstructed networks and we validated the relevance of the microfluidic platform in the context of Huntington’s disease. We then studied the evolution of intracellular dynamics with the maturation of reconstructed neuronal networks in microfluidic chambers. We observed an increase of anterograde axonal transport of secretory vesicles during maturation. These first results lead us to think that neuronal activity could regulate axonal transport of secretory vesicles over maturation of the network.Therefore, we improved the in vitro microfluidic system with a designed microelectrode array (MEA) substrate allowing us to record intracellular dynamics while controlling neuronal activity. Using this system, we identified an axon-resident reserve pool of secretory vesicles recruited upon neuronal activity to rapidly distribute secretory materials to presynaptic sites. We identified the activity-dependent mechanism of recruitment of this axonal pool of vesicles along the axon shaft. We showed that Myosin Va ensures the tethering of vesicles in the axon shaft in axonal actin structures. Specifically, neuronal activity induces a calcium increase after activation of Voltage Gated Calcium Channels along the axon, which regulates Myosin Va and triggers the recruitment of tethered vesicles on microtubules. We then showed the involvement of this activity-dependent pool for axon branches formation during axon development. By developing 2-photon live microscopy of axonal transport in acute slices, we finally confirmed that a pool of axon-resident static vesicles is recruited by neuronal activity in vivo with a similar kinetic.Altogether, this work provides new in vitro and in vivo tools to study intracellular dynamics in physiological networks. Using these tools, we identified the existence of a local mechanism of axonal transport regulation along the axon shaft, allowing rapid supply of trophic factors to developing branches

Abstract:

Les ARNms des cellules eucaryotes sont liés à des protéines de liaison aux ARNs (RBPs) et empaquetés au sein d’assemblages macro-moléculaires appelés granules RNP. Dans les cellules neuronales, les granules RNP de transport sont impliqués dans le transport d’ARNms spécifiques jusqu’aux axones et dendrites, ainsi que dans leur traduction locale en réponse à des signaux externes. Bien que peu de choses soient connues sur l’assemblage et la régulation de ces granules in vivo, des résultats récents ont indiqué que la présence de domaines de type prion (PLDs) dans les RBPs facilite les interactions protéines-protéines et protéines-ARN, favorisant ainsi la condensation de complexes solubles en granules RNP. La RBP conservée Imp est un composant central de granules RNP qui sont transportés dans les axones lors du remodelage neuronal chez la drosophile. De plus, la fonction de Imp est nécessaire au remodelage des axones lors de la maturation du système nerveux de drosophile. Une analyse de la séquence de la protéine Imp a révélé qu’en plus de quatre domaines de liaison aux ARNs, Imp contient un domaine C-terminal désordonné enrichi en Glutamines et Serines, deux propriétés caractéristiques des domaines PLDs. Lors de ma thèse, j’ai étudié la fonction de ce PLD dans le contexte de l’assemblage et du transport des granules RNP. J’ai observé en culture de cellules que les granules Imp s’assemblent en absence de PLD, bien que leur nombre et leur taille soient augmentés. Des protéines présentant une séquence PLD mélangée, au contraire, s’accumulent dans des granules au nombre et à la taille normale, indiquant que l’état désordonné de ce domaine, et non sa séquence primaire, est essentiel à l’homéostasie des granules. De plus, des expériences de FRAP réalisées en culture de cellule et in vivo ont révélé que le domaine PLD de Imp favorise la dynamique des granules. In vivo, ce domaine est nécessaire et suffisant à l’accumulation axonale de Imp. Comme montré par une analyse en temps réel, l’absence de domaine PLD aboutit également à une diminution du nombre de granules axonaux motiles. Fonctionnellement, le domaine PLD de Imp est essentiel au remodelage neuronal car des protéines sans ce domaine ne sont pas capables de supprimer les défauts de repousse axonale observés après inactivation de imp. Enfin, la génération d’un variant de Imp dans lequel le domaine PLD a été déplacé en N-terminus a montré que les fonctions du PLD dans le transport des granules et dans leur assemblage sont découplées, et que la modulation des propriétés des granules Imp médiée par le domaine PLD n’est pas nécessaire au remodelage neuronal in vivo. En conclusion, mes résultats ont montré que le domaine PLD de Imp n’est pas nécessaire à l’assemblage des granules RNP Imp, mais régule leur nombre et leur dynamique. De plus, mon travail a mis en évidence une fonction inattendue pour un domaine PLD dans le transport axonal et le remodelage des neurones lors de la maturation du système nerveux Eukaryotic mRNAs are bound by RNA Binding Proteins (RBP) and packaged into diverse range of macromolecular assemblies named RNP granules. In neurons, transport RNP granules are implicated in the transport of specific mRNAs to axons or dendrites, and in their local translation in response to external cues. Although little is known about the assembly and regulation of these granules in vivo, growing evidence indicates that the presence of Prion Like domains (PLD) within RBPs favours multivalent protein–protein and protein-RNA interactions, promoting the transition of soluble complexes into RNP granules. The conserved RBP Imp is as a core component of RNP granules that are actively transported to axons upon neuronal remodelling in Drosophila. Furthermore, Imp function was shown to be required for axonal remodelling during Drosophila nervous system maturation. Analyses of the domain architecture of the Imp protein revealed that, in addition to four RNA binding domains (RBD), Imp contains a Cterminal domain showing a striking enrichment in Glutamines and Serines, which is one of the characteristics of a PLD. During my PhD, I explored the function of the PLD in the context of granule assembly and transport. In cultured cells, I observed that Imp granules assembled in the absence of the PLD, however their number and size were increased. Proteins with scrambled PLD sequence accumulated in granules of normal size and number, implying that the degree of disorder of this domain, and not its sequence, is essential for granule homeostasis. Moreover, FRAP experiments, performed on cultured cells and in vivo, revealed that Imp PLD is important to maintain the turnover of these granules. In vivo, this domain is both necessary and sufficient for efficient transport of Imp granules to axons. These defects are associated with a reduction on the number of motile granules in axons. Furthermore, mutant forms lacking the PLD do not rescue the axon remodelling defects observed upon imp loss of function. Finally, a swapping experiment in which I moved Imp PLD from the C-terminus to the N-terminus of the protein revealed that the functions of Imp PLD in granule transport and homeostasis are uncoupled, and that PLD-dependent modulation of Imp granule properties is dispensable in vivo. Together, my results show that Imp PLD of is not required for the assembly of RNP granules, but rather regulates granule number and dynamics. Furthermore, my work uncovered an unexpected in vivo function for a PLD in axonal transport and remodelling during nervous system maturation

Le développement du cortex cérébral se déroule selon des étapes bien définies qui sont essentielles à la formation d’un cerveau fonctionnel. La perturbation de l’une ou plusieurs de ces étapes peut conduire à des malformations neuro-développementales, responsables de différents troubles cognitifs, d’épilepsies ou encore de déficience intellectuelle. De nombreuses mutations dans des gènes codant pour les tubulines ou bien les kinésines, sont retrouvées chez des individus présentant diverses anomalies neuro-développementales. Bien que les kinésines soient impliquées dans le développement cortical, les mécanismes fonctionnels par lesquels elles conduisent aux malformations demeurent encore méconnus. Mon travail de thèse identifie la kinésine Kif21b, jusqu’alors peu connue, comme étant essentielle au développement cortical. Nous montrons que Kif21b régule la migration neuronale dans le cortex et identifions quatre variants chez des individus présentant des malformations neuro-développementales. Nous montrons que l’expression ectopique des variants chez la souris et le poisson zèbre récapitulent les phénotypes observés chez ces patients The development of the cerebral cortex is a highly regulated process that is crucial for the establishment of functional cortical networks. Disruption of one or several of these steps can lead severe neurodevelopmental disorders that are associated with intellectual disabilities, epilepsies and cognitive impairment. Over the past few years, several genetic mutations in genes encoding either tubulin or microtubule-associated motors such as kinesins, have been found in individuals with neurodevelopmental disorders. Although kinesins have been found to be essential for a proper cortical development, the exact functions of kinesins in these processes are still poorly understood. My work clearly identified Kif21b, a poorly-known kinesin, as a novel key regulator of cortical development both in mouse and human. We show that Kif21b regulates both radial and tangential migration of cortical neurons, and identify four KIF21B variants in individuals presenting neurodevelopmental disorders. We show that ectopic expression of variants recapitulate phenotypes both in mice and zebrafish

Abstract:

Amyotrophic Lateral Sclerosis (ALS) is a rare and fatal neurodegenerative disorder resulting in the loss of motor neurons from the spinal cord and frontal cortex. The patterns of neurodegeneration, affected regions, age of onset, and time course of disease progression are all highly variable between and within variants of the disease. Familial ALS (fALS), inherited versions of ALS due to genetic changes, accounts for between 5-20% of all ALS cases, while the rest are sporadic, with either no causative mutation identified or no familial history of ALS. Recently, the discovery of C9orf72 hexanucleotide repeat expansions have been identified as one of the most common causes of familial ALS, with some patients presenting with dual phenotypes of ALS and frontotemporal dementia, leading to new hypotheses about the nature of neurodegenerative diseases. Despite the continued discovery of new ALS causative genes, little is known about the pathogenesis of the disease. While almost all variants include the presence of intracellular protein inclusions, the site of the protein plaques and involved proteins varies between genetic and phenotypic variants of this disease. Due to the lack of clear pathogenic mechanisms, several hypotheses have been developed to explain the process of neurodegeneration. Autophagy, the process of self-eating, leading to destruction of damaged or excess proteins and organelles, has been implicated as being altered in ALS. Multiple variants have demonstrated altered mitochondrial morphology and cellular energetic dynamics, which could explain previous observations that implicate the process of apoptosis in cellular death. Many of the involved proteins in ALS have functional roles for intracellular, nucleocytoplasmic, and axonal transport of various proteins or RNA. These three competing hypotheses are currently the most prominent hypotheses in the pathogenesis of ALS, and have largely been considered as separate and competing in past research. Is there a chance that the true pathogenesis leading to neuronal destruction via apoptosis involve all three hypotheses? Altered protein and RNA transport dynamics could lead to changes in cellular stress responses or overload autophagy pathways, leading to exacerbated cellular stress responses, leading to alterations in mitochondrial morphology and eventually cell death via apoptosis.

The central event driving neuronal activity is represented by synaptic transmission, a process that relies on regulated cycles of synaptic vesicle (SV) exocytosis and endocytosis at presynaptic terminal level. Neurons, polarized and perennial cells, to guarantee an efficient neurotransmitter release, to maintain cellular homeostasis and promote neuronal survival, are particularly dependent on efficient quality control pathways to continuously remove dysfunctional presynaptic proteins and organelles. The main mechanisms used by neurons to achieve these goals are endosomal sorting and autophagy, a highly conserved endo-lysosomal degradation pathway required to recycle basic nutrients by the clearance of damaged or aged proteins and organelles. Several presynaptic endocytic proteins have been shown to regulate both SV recycling and autophagy and defects in both pathways have been linked to neurodevelopmental abnormalities and neurodegeneration in mouse and humans. In 2017 we characterized the previously unknown protein APache (KIAA1107) as a neuronal-specific protein, novel interactor of the adaptor protein AP-2 essential in the regulation of neuronal development and SV cycle in vitro and in vivo. In this work, we intended to define APache functional role in neuronal autophagy by combining electron microscopy, immunofluorescence, live-cell imaging microscopy and biochemistry. We observed that APache is actually involved in autophagy: the induction of the process increases APache levels in mature neurons and, conversely, APache silencing leads to a severe accumulation of late-stage autophagosomes in neurons, also at synaptic level, due to autophagic blockade. Interestingly, APache expression is significantly reduced in the brain of sporadic Alzheimer’s disease patients. These data point to APache as a novel key regulator of neuronal autophagy. Its altered levels, resulting in defective autophagy, may contribute to the precocious cellular alterations and synaptic dysfunctions observed in neurodegenerative diseases. Abstract:

Ce travail introduit une nouvelle methode d'interpretation de la vision du monde reel et applique cette technique a la navigation d'un robot mobile autonome dans un milieu industriel. Stan, systeme de transport a apprentissage neuronal, a pour objectif d'accroitre a moindre cout, la flexibilite et l'autonomie locale de robots mobiles. Cette proposition s'appuie essentiellement sur deux points. Premierement, le systeme de perception, base sur un systeme de vision omnidirectionnelle, permet de collecter des informations dans toutes les directions autour du robot en une seule prise d'image. Ce systeme consiste en la photographie du reflet de l'environnement sur un miroir conique. L'image obtenue fournit une signature caracteristique de la position dans l'environnement. Cette methode de perception est etudiee a travers deux prototypes: l'un permettant la prise d'une seule image, l'autre realisant la prise d'un couple d'images stereoscopiques. Deuxiemement, notre point de vue revise les developpements actuels des methodes de guidage et de navigation. Il s'appuie sur la reconnaissance des signatures caracteristiques de la position que fournit le systeme de perception afin de calculer la position du vehicule. La methode se decompose en deux etapes: un mode apprentissage ou un operateur enseigne la trajectoire de travail au robot, et, un mode operatoire ou le robot realise une reconnaissance des images recues. Ces deux phases constituent les etapes d'entrainement et d'utilisation de reseaux de neurones a retropropagation du gradient. Ce memoire souligne dans un premier temps la problematique liee a l'utilisation de vehicules autonomes dans un milieu industriel. Ces observations amenent alors la proposition d'une solution originale dont les differents points specifiques sont ensuite abordes a travers une presentation generale du projet stan. Un ensemble de resultats issus de simulations et experimentations permet ensuite la validation de l'idee generale a travers la comparaison de plusieurs architectures neuronales. Un bilan des contributions et des perspectives ouvertes par le projet stan conclut enfin sur l'avancement des travaux

Présenté initialement à l'agence spatiale américaine par P. Glaser comme une source potentielle d'énergie alternative renouvelable et propre, le projet de Centrales Solaires Orbitales est basé sur le concept de Transport d'Energie Sans Fil (TESF). Le principe consiste à collecter directement dans l'espace l'énergie solaire, puis à la transmettre vers une base de réception terrestre, via un faisceau hyperfréquence. Dans le cadre des actions menées dans le domaine du TESF au niveau terrestre, une solution technologique permettant de répondre au cahier des charges imposé au système d'émission a retenu notre attention : mettre en oeuvre un réseau d'antenne phasé alimenté par des magnétrons de moyenne puissance. Dans cette optique, ce travail de recherche présente une approche originale du contrôle des grandeurs de sortie d'un magnétron opérant en situation de verrouillage par injection. Afin de prendre en compte le comportement non linéaire du magnétron, une stratégie de contrôle hybride a été mise en oeuvre pour le contrôle de la fréquence et de l'amplitude d'un magnétron de moyenne puissance (2.45 GHz) verrouillé par injection et débitant sur une charge fixe. L'aspect hybride est constitué par l'association d'un algorithme de Contrôle Direct Inverse impliquant un réseau de neurones non linéaires modélisant la fonction de transfert inverse du magnétron, avec un correcteur linéaire en boucle fermée de type PID. Le développement d'un dispositif de caractérisation expérimentale d'un magnétron verrouillé par injection a permis de collecter des bases de mesures nécessaires à un apprentissage supervisé et généralisé pour l'identification du contrôleur neuronal. Les meilleures performances en terme de conduite du magnétron ont été obtenues avec une boucle de contrôle effectuant une permutation dynamique entre le correcteur neuronal non linéaire et le correcteur linéaire PID tout en assurant une stabilité de la phase sur l'ensemble de la bande de verrouillage

Abstract:

Le transporteur neuronal de la dopamine (TDA), un élément fonctionnel impliqué dans la régulation synaptique de la dopamine (DA), constitue la cible moléculaire des drogues comme la cocaïne et ses dérivés. Le but du présent travail est d'élucider les mécanismes impliqués dans le fonctionnement du TDA, en vue d'identifier le type de molécule capable de s'opposer à la liaison des drogues sur ce dernier. Pour ce faire, nous avons caractérisé in vitro et in vivo le pouvoir inhibiteur irréversible du DEEP-NCS et l'avons utilisé pour mesurer la vitesse de renouvellement du TDA, qui est d'environ 6 jours. Il n'y a pas de modification du contenu en ARNm du TDA au décours de son inactivation par le DEEP-NCS. Ces résultats démontrent que la vitesse de renouvellement du TDA mesurée dans la présente étude pourrait être celle qui prévaut dans les conditions physiologiques. Nous avons comparé par la suite la capacité des inhibiteurs et des substrats du TDA à protéger la liaison d'un analogue de la cocaïne, le WIN 35,428, vis-à-vis des effets alkylants du N-éthylmaléimide, et avons montré qu'il existe une distinction entre les sites de liaison de ces deux groupes d'agents pharmacologiques. Enfin, nous avons déterminé le mode de liaison d'un inhibiteur spécifique et puissant du TDA, le GBR 12783, un agent dont la liaison sur le TDA s'oppose à celle de la cocaïne. Nous avons donc pour la première fois montré que le GBR 12783 se lie sur le TDA en deux étapes consécutives, avec une deuxième étape qui conduit lentement à la formation d'un complexe TDA-GBR 12783 stable. Le GBR 12783 constitue donc le prototype à partir duquel des agents bloquant la liaison de la cocaïne sur sa cible moléculaire pourrait être développés. En conclusion, ce travail fournit des informations sur les données fonctionnelles et pharmacologiques du TDA, pouvant aider à la conception de produits efficaces contre la cocaïnomanie.

Abstract:

Les mitochondries assurent des fonctions essentielles dans les cellules via la production d'énergie sous forme d'ATP, la captation de calcium et la régulation de la mort cellulaire par apoptose. Les dysfonctions des mitochondries apparaissent à des stades précoces de la maladie d'Alzheimer (MA), et ont été particulièrement associées au peptide toxique Aβ. Ce dernier est issu du clivage séquentiel de la protéine précurseur de l'amyloïde (APP) par la β- et la γ-sécrétase. Plusieurs traitements ciblant l'Aβ se sont avérés inefficaces contre la progression de la MA, orientant les recherches sur le potentiel toxique d'autres fragments issus du clivage de l'APP. L'hypothèse de mon projet porte sur la toxicité spécifique des fragments APP C-terminaux (APP-CTFs : C83 et C99 (précurseur direct de l'Aβ)) en se focalisant sur l'étude de la structure, la fonction des mitochondries et la mitophagie. J'ai également étudié l'impact de l'APP et de ses fragments sur la machinerie de transport des mitochondries, un mécanisme clé de leur renouvellement, en particulier dans les neurones.Premier axe : Impact de l'accumulation des APP-CTFs sur la structure, la fonction des mitochondries et sur la mitophagie. Nous décrivons une accumulation des APP-CTFs dans la fraction mitochondriale de modèles mimant les formes familiales de la MA in vitro (cellules de neuroblastome humains exprimant l'APP portant la double mutation suédoise (SH-SY5Y-APPswe), ou le fragment C99 (SH-SY5Y-C99)) et in vivo (souris transgéniques 3xTgAD exprimant les mutations APPswe, TauP301L, PS1 (M146V) ou bien le fragment C99 après infection virale). Par le biais d'une approche pharmacologique bloquant l'activité de la γ-sécrétase, nous démontrons in vitro et in vivo que l'accumulation des APP-CTFs, indépendamment de l'Aβ, est associée à l'agglomération de mitochondries structurellement altérées et surproduisant des espèces oxygénées réactives toxiques, conjointement à un blocage de la mitophagie. Nous avons conclu notre étude par la démonstration de l'altération de la mitophagie dans les cerveaux de patients atteints de la MA sporadique, corrélant avec les niveaux d'APP-CTFs (1, 2).Second axe : Etude des effets de l'APP, des APP-CTFS et de l'Aβ sur les protéines de transport mitochondrial. J'ai d'abord démontré l'impact de l'APP endogène et de la surexpression de l'APPswe sur les niveaux des protéines de la machinerie de transport mitochondrial in vitro (fibroblastes de souris KO pour l'APP ainsi que dans cellules SH-SY5Y-APPswe). J'ai discriminé le rôle de l'accumulation des APP-CTFs de celui de l'Aβ sur l'expression de ces protéines en traitant les cellules APPswe avec l'inhibiteur de la γ-sécrétase. J'ai validé ces observations en utilisant des fibroblastes de souris déplétés des présénilines (composants catalytiques de la γ-sécrétase) mimant une accumulation des APP-CTFs. J'ai par ailleurs démontré une implication des APP-CTFs et de l'Aβ dans le défaut de recrutement des mitochondries à la machinerie de transport dans les cellules SHSY-5Y différentiées. En analysant des cerveaux de souris 3xTgAD et WT à différent âges et des échantillons de cerveaux de patients Alzheimer sporadiques à différents stades de la maladie, nous rapportons que le développement de la MA et l'âge en lui-même ont un impact différentiel sur l'expression de certaines protéines de transport mitochondrial (3).Ces études démontrent de nouveaux mécanismes moléculaires impactant l'homéostasie mitochondriale au cours du développement de la MA. These functions are altered at early stages of Alzheimer's disease (AD) and are essentially induced by the Amyloid (Aβ), produced after the sequential cleavage of amyloid precursor protein (APP) by β- and γ-secretase. Aβ is a major actor of AD development but all the treatments targeting this peptide remain ineffective. C-terminal APP fragments (APP-CTFs: C83 and C99 (Aβ precursor) are other fragments presenting specific toxicity in AD and new potential therapeutic targets. My project is focus on the study of APP-CTFs toxicity, independently of Aβ, on the structure, function of mitochondria, their degradation by mitophagy and on mitochondrial transport proteins. They constitute the complex allowing mitochondrial transport in cells, especially in neurons, closely linked to mitochondrial renewal, particularly in neurons.First axe: APP-CTFs impact on mitochondrial structure, function and mitophagy. We described APP-CTFs accumulation in mitochondrial fraction in vitro (human neuroblastoma cells expressing APP Swedish double mutation (SH-SY5Y-APPswe) or C99 fragment (SH-SY5Y-C99)) and in vivo (3xTgAD mice expressing APPswe, TauP301L, PS1 (M146V) or C99 fragment after viral injection). We inhibit the cleavage of APP-CTFs and the production of Aβ by pharmacological approaches, to abolish γ-secretase activity. Ours results show for the first time in vitro and in vivo, that high concentration of APP-CTFs independently of Aβ, impact mitochondrial structure, function and alter mitophagy process, resulting in an accumulation of altered mitochondria producing high levels of toxic reactive oxygen species. In addition, our results in patient brains of sporadic AD (SAD) patients show altered mitophagic protein levels correlating with APP-CTFs accumulation (1-2).Second axe: study of the effects of APP, APP-CTFs and Aβ peptide on mitochondrial transport machinery. I reported the specific regulation of mitochondrial transport protein by endogenous APP (Mice fibroblasts APP WT and KO) and the overexpression of APPswe (and in SH-SY-5Y-APPswe cells). APP-CTFs and Aβ differentially regulate mitochondrial transport protein levels in treated SH-SY-5Y-APPswe cells with γ-secretase inhibitor. These results were validated in mice fibroblasts KO for presenilins (catalytic compounds of γ-secretase) avoiding APP-CTFs degradation. APP-CTFs and Aβ impair the recruitment of mitochondria to its transport machinery in differentiated SHSY-5Y. The progression of the disease deregulates the levels of mitochondrial transport protein in vivo (3xTgAD and WT mice brains, C99 injected mice brains) and in SAD patients brains. The analyses of young and old mice brains and of SAD patients samples at different stages of the disease, allowed us to demonstrate an impact of aging in the regulation of mitochondrial transport protein levels. This phenomenon occurs also in addition with AD progression (3).These studies highlight new molecular mechanisms impacting mitochondrial homeostasis during AD progression. Abstract:

Endocytic trafficking dynamically regulates neuronal plasma membrane protein presentation and activity, and plays a central role in excitability and plasticity. Over the course of my dissertation research I investigated endocytic mechanisms regulating two neuronal membrane proteins: the anesthetic-activated potassium leak channel, KCNK3, as well as the psychostimulant-sensitive dopamine transporter (DAT). My results indicate that KCNK3 internalizes in response to Protein Kinase C (PKC) activation, using a novel pathway that requires the phosphoserine binding protein, 14-3-3β, and demonstrates for the first time regulated KCNK3 channel trafficking in neurons. Additionally, PKC-mediated KCNK3 trafficking requires a non-canonical endocytic motif, which is shared exclusively between KCNK3 and sodium-dependent neurotransmitter transporters, such as DAT. DAT trafficking studies in intact ex vivo adult striatal slices indicate that DAT endocytic trafficking has both dynamin-dependent and –independent components. Moreover, DAT segregates into two populations at the neuronal plasma membrane: trafficking-competent and -incompetent. Taken together, these results demonstrate that novel, non-classical endocytic mechanisms dynamically control the plasma membrane presentation of these two important neuronal proteins.

Abstract:

Les ARNms des cellules eucaryotes sont liés à des protéines de liaison aux ARNs (RBPs) et empaquetés au sein d’assemblages macro-moléculaires appelés granules RNP. Dans les cellules neuronales, les granules RNP de transport sont impliqués dans le transport d’ARNms spécifiques jusqu’aux axones et dendrites, ainsi que dans leur traduction locale en réponse à des signaux externes. Bien que peu de choses soient connues sur l’assemblage et la régulation de ces granules in vivo, des résultats récents ont indiqué que la présence de domaines de type prion (PLDs) dans les RBPs facilite les interactions protéines-protéines et protéines-ARN, favorisant ainsi la condensation de complexes solubles en granules RNP. La RBP conservée Imp est un composant central de granules RNP qui sont transportés dans les axones lors du remodelage neuronal chez la drosophile. De plus, la fonction de Imp est nécessaire au remodelage des axones lors de la maturation du système nerveux de drosophile. Une analyse de la séquence de la protéine Imp a révélé qu’en plus de quatre domaines de liaison aux ARNs, Imp contient un domaine C-terminal désordonné enrichi en Glutamines et Serines, deux propriétés caractéristiques des domaines PLDs. Lors de ma thèse, j’ai étudié la fonction de ce PLD dans le contexte de l’assemblage et du transport des granules RNP. J’ai observé en culture de cellules que les granules Imp s’assemblent en absence de PLD, bien que leur nombre et leur taille soient augmentés. Des protéines présentant une séquence PLD mélangée, au contraire, s’accumulent dans des granules au nombre et à la taille normale, indiquant que l’état désordonné de ce domaine, et non sa séquence primaire, est essentiel à l’homéostasie des granules. De plus, des expériences de FRAP réalisées en culture de cellule et in vivo ont révélé que le domaine PLD de Imp favorise la dynamique des granules. In vivo, ce domaine est nécessaire et suffisant à l’accumulation axonale de Imp. Comme montré par une analyse en temps réel, l’absence de domaine PLD aboutit également à une diminution du nombre de granules axonaux motiles. Fonctionnellement, le domaine PLD de Imp est essentiel au remodelage neuronal car des protéines sans ce domaine ne sont pas capables de supprimer les défauts de repousse axonale observés après inactivation de imp. Enfin, la génération d’un variant de Imp dans lequel le domaine PLD a été déplacé en N-terminus a montré que les fonctions du PLD dans le transport des granules et dans leur assemblage sont découplées, et que la modulation des propriétés des granules Imp médiée par le domaine PLD n’est pas nécessaire au remodelage neuronal in vivo. En conclusion, mes résultats ont montré que le domaine PLD de Imp n’est pas nécessaire à l’assemblage des granules RNP Imp, mais régule leur nombre et leur dynamique. De plus, mon travail a mis en évidence une fonction inattendue pour un domaine PLD dans le transport axonal et le remodelage des neurones lors de la maturation du système nerveux Eukaryotic mRNAs are bound by RNA Binding Proteins (RBP) and packaged into diverse range of macromolecular assemblies named RNP granules. In neurons, transport RNP granules are implicated in the transport of specific mRNAs to axons or dendrites, and in their local translation in response to external cues. Although little is known about the assembly and regulation of these granules in vivo, growing evidence indicates that the presence of Prion Like domains (PLD) within RBPs favours multivalent protein–protein and protein-RNA interactions, promoting the transition of soluble complexes into RNP granules. The conserved RBP Imp is as a core component of RNP granules that are actively transported to axons upon neuronal remodelling in Drosophila. Furthermore, Imp function was shown to be required for axonal remodelling during Drosophila nervous system maturation. Analyses of the domain architecture of the Imp protein revealed that, in addition to four RNA binding domains (RBD), Imp contains a Cterminal domain showing a striking enrichment in Glutamines and Serines, which is one of the characteristics of a PLD. During my PhD, I explored the function of the PLD in the context of granule assembly and transport. In cultured cells, I observed that Imp granules assembled in the absence of the PLD, however their number and size were increased. Proteins with scrambled PLD sequence accumulated in granules of normal size and number, implying that the degree of disorder of this domain, and not its sequence, is essential for granule homeostasis. Moreover, FRAP experiments, performed on cultured cells and in vivo, revealed that Imp PLD is important to maintain the turnover of these granules. In vivo, this domain is both necessary and sufficient for efficient transport of Imp granules to axons. These defects are associated with a reduction on the number of motile granules in axons. Furthermore, mutant forms lacking the PLD do not rescue the axon remodelling defects observed upon imp loss of function. Finally, a swapping experiment in which I moved Imp PLD from the C-terminus to the N-terminus of the protein revealed that the functions of Imp PLD in granule transport and homeostasis are uncoupled, and that PLD-dependent modulation of Imp granule properties is dispensable in vivo. Together, my results show that Imp PLD of is not required for the assembly of RNP granules, but rather regulates granule number and dynamics. Furthermore, my work uncovered an unexpected in vivo function for a PLD in axonal transport and remodelling during nervous system maturation

Les circuits neuronaux régissent les comportements tels que la coordination motrice ou la mémoire et l’apprentissage. Au sein d’un réseau, les neurones communiquent par des processus moléculaires et cellulaires finement réglés à la synapse. Un des mécanismes régulant l’homéostasie synaptique, le transport de vésicules dans les neurones, est dérégulé dans les maladies neurologiques telles que le syndrome de Rett, la maladie d’Alzheimer et la maladie d’Huntington. Ainsi, investiguer la régulation du transport de vésicules dans les neurites dans un contexte physiologique est important pour comprendre, et potentiellement rétablir, les conséquences de ces dérégulations pathologiques.La protéine Huntingtine (HTT), connue pour son implication dans la maladie d’Huntington, est un acteur clé du transport axonal. Elle promeut et influence le transport des vésicules en favorisant le recrutement des adaptateurs et des moteurs moléculaires. Sa phosphorylation sur la sérine 421 (pHTTS421) régule la directionnalité des vésicules de BDNF, d’APP et de VAMP-7 dans des neurones transfectés in vitro. Cependant, les mécanismes et les conséquences de la régulation du transport par HTT comme la spécificité neuritique et les conséquences comportementales restent peu connus. Enfin, nous ignorons si la régulation du transport peut être influencée dans des conditions pathologiques afin de restaurer les phénotypes in vivo.Ce projet de thèse vise à caractériser les mécanismes et les conséquences de la régulation du transport axonal de trois types de vésicules par pHTTS421 et d’investiguer sa propension à restaurer les phénotypes associés à des maladies neurologiques dans des modèles de souris.Dans le but de reproduire in vitro les réseaux associés à des maladies neurologiques, nous avons utilisé des chambres microfluidiques. Nous avons étudié le transport des vésicules d’APP, des précurseurs des vésicules synaptiques (PVSs) ou des vésicules à cœur dense (VCDs) contenant BDNF au sein d’un réseau neuronal dans lequel pHTTS421 a été modifiée. Ces neurones sont issus de souris pour lesquelles la sérine 421 a été remplacée par un acide aspartique ou par une alanine pour mimer respectivement l’état phosphorylé (HTTS421D) ou non phosphorylable (HTTS421A) de la HTT.Dans la maladie d’Alzheimer, l’homéostasie d’APP est dérégulée. Nous avons donc étudié son transport et son accumulation synaptique dans un circuit corticocortical. Nous avons trouvé que la phosphorylation de la sérine S421 par Akt régule la directionnalité des vésicules d’APP uniquement dans les axones : HTTS421A diminue le flux antérograde axonal d’APP ainsi que ses niveaux à la synapse in vitro et in vivo. Réduire le flux antérograde d’APP dans un modèle murin d’Alzheimer restaure l’homéostasie synaptique in vivo et les déficits de mémoire associés (publication 1 ; Bruyère*, Abada*, Vitet* et al., eLife, 2020).Le transport axonal des PVSs régule le nombre de vésicules synaptiques (VSs), ce qui, dans un réseau corticostriatal, est essentiel à l’apprentissage de compétences motrices. Nous avons montré que pHTTS421 augmente le recrutement de la kinésine KIF1A sur les PVSs, augmentant le transport antérograde et la probabilité d’exocytose. En réduisant les niveaux de KIF1A dans le réseau corticostriatal des souris HTTS421D, nous avons trouvé que pHTTS421 augmente le nombre de VSs et altère la mémoire procédurale. Cette étude décrit comment le transport axonal des PVSs impacte les phénotypes comportementaux (publication 2 ; vitet et al., in prep).Enfin, le transport de BDNF est dérégulé dans le réseau corticostriatal des jeunes filles atteintes du syndrome de Rett. Nous avons observé que l’expression endogène de pHTTS421 ou l’injection d’un composé inhibant la calcineurine (FK506) restaure le transport de BDNF dans un réseau corticostriatal, la communication neuronale et les symptômes associés chez les souris modèles du syndrome de Rett (Publication 3 ; Ehinger et al., Embo Mol Med, 2020) Neuronal circuits are at the basis of behaviors such as motor coordination or learning and memory. As being part of a network, neurons communicate at synapses through finely tuned molecular and cellular processes. One key mechanism regulating synapse homeostasis involves transport of vesicles within axons and dendrites which is dysregulated in many neurological disorders such as Rett syndrome, Alzheimer’s (AD) and Huntington’s diseases (HD). Thus, deciphering the regulation of vesicular transport within neurites in physiological context is crucial to understand, and potentially restore, the consequences of these dysregulations in pathological contexts.Huntingtin (HTT) protein, known for its devastating role in HD when mutated, is a key actor of axonal transport. It promotes and regulates vesicular transport in neurites by scaffolding adaptors and molecular motors. Particularly, HTT phosphorylation status on S421 regulates the directionality of BDNF, APP and VAMP-7 vesicles within neurites in cultured and transfected neurons. However, several questions remain to be elucidated regarding the mechanisms and the consequences of this HTT-dependent regulation of axonal transport such as the neuritic specificity (axons or dendrites) and the behavioral consequences of such modifications. Finally, we do not know whether transport regulation can be influenced in pathological conditions to restore disease-associated phenotypes in vivo.This thesis aims at characterizing in vivo the mechanisms and the consequences of axonal transport regulation of three different vesicles through the phosphorylation of Huntingtin at S421 and to investigate its propensity to restore disease-associated phenotypes in mouse models of human neurological disorders.In order to reproduce in vitro the in vivo networks associated to neurological disorders we used microfluidic devices. We investigated the transport of Amyloid Precursor Protein (APP) vesicles, precursors of synaptic vesicles (SVPs) or dense-core vesicles (DCVs) in neurons in which the HTT phosphorylation status was modified. These neurons came from mice in which Serine 421 has been replaced by an aspartic acid to mimic the phosphorylated form of HTT (HTTS421D) or by an alanine to mimic the unphosphorylatable form of HTT (HTTS421A).Transport of APP vesicles is impaired in AD. We investigated APP transport and accumulation at synapses within a corticocortical circuit. We found that Akt-mediated HTT phosphorylation at S421 regulates the directionality of APP containing vesicles in axons but not in dendrites: the phospho-defective form of HTT decreases axonal anterograde flux of APP and reduces its levels at presynaptic zones both in vitro and in vivo. Reducing anterograde flux of APP in familial AD mouse model restored synapse homeostasis in vivo and memory deficits (Publication 21; Bruyere*, Abada*, Vitet* et al., eLife, 2020).SVP axonal transport regulates the number of SVs at the synapse, which, within a corticostriatal synapse, is essential for motor skill learning. We found that HTT phosphorylation increases the recruitment of the molecular motor KIF1A on SVPs, thus promoting anterograde transport and the probability of release. Silencing KIF1A in the corticostriatal network of HTTS421D mice, we found that pHTTS421 increases the number of SVs at the synapse and impairs procedural memory through a specific HTT-KIF1A dependent mechanism. This study defines a pathway by which axonal transport of SVP impact the behavioral phenotype. (Publication 2; Vitet et al, in prep)Finally, it has been shown that BDNF transport within DCVs is dysregulated in the corticostriatal network of Rett syndrome’s patients. En vue de comprendre les rapports étroits qui existent entre les gradients électrochimiques transmembranaires et le processus de capture, nous avons étudié le rôle du KC1 dans la NA +-dépendance de la capture de la [3H]dopamine (DA) réalisée par des synaptosomes issus du striatum de rat. Nous avons ainsi montré que la capture est optimale lorsque les concentrations de Na + et de K + physiologiques extracellulaires sont reproduites. Leurs modifications provoquent un blocage de la capture de [3H]DA résultant de la perturbation de l'activité de l'ATPase Na +-K +-dépendante qui ne peut plus assurer le maintien des gradients ioniques transmembranaires. Nous avons ensuite tenté de caractériser la nature des interactions entre les ions (Na +, K +, Ca 2 +,) et la liaison au transporteur neuronal de la DA (TDA) d'un analogue de la cocaïne, le [3H]WIN 35428. Les résultats révèlent l'existence au sein du TDA de 2 sites par lesquels les ions régulent la liaison du [3H]WIN 35428 : un site externe et un site interne situés respectivement à l'extérieur et à l'intérieur du domaine de liaison du radioligand. Les sites externe et interne seraient liés par des interactions allostériques négatives alors que le site externe nous avons comparé l'effet des ions sur les liaisons d'inhibiteurs et de substrats au TDA marqué avec le [3H]Mazindol. Les résultats obtenus permettent de proposer un modèle du TDA qui comporterait des sites de liaison différents mais qui se recouvriraient partiellement. En extrapolant ces données à la capture de la DA, nous avons émis un modèle de fonctionnement du TDA dans le processus de capture dans les conditions physiologiques.

Nous avons effectué une étude comparative du transport de la [3H] DA et de la liaison spécifique de [3H] GBR 12783 au transporteur, en utilisant des préparations obtenues à partir de striatum de rat. La dépendance des constantes cinétiques du processus de capture vis-à-vis de la concentration des ions Cl est compatible avec un cotransport DA-Cl. La dépendance de la liaison des substrats au site marque par le [3H] GBR 127833 vis-à-vis du Cl est d'autant plus importante que le substrat a une affinité marquée pour le transporteur. Comme pour la liaison de [3H] GBR 12783 aucun cation n'est capable de substituer des ions Na+ sans provoquer par lui-même une inhibition de la capture de [3H] DA. Les profils comparés de Na+ dépendance de la capture neuronale de [3H] DA et de la liaison spécifique de [3H] GBR 12783révèlent l'effet inhibiteur de certains cations (K+, Ca++, Mg++) sur les deux processus et indiquent que ce pouvoir est d'autant plus important que la concentration de Na+ est faible. La liaison des substrats nécessiterait qu'une faible concentration de Na+ (3 ) 5 mM). La liaison spécifique de [3H] GBR 12783 et la capture spécifique de [3H] DA présentent la même sensibilité vis-à-vis des ions métalliques tels que (Hg++, Cd++, Cu++), suggérant l'implication des groupements -SH dans ces deux processus. Les groupements -SH qui lient le Zn++ joueraient un rôle important dans la translocation de la DA

Abstract:

G proteins transduce a myriad of signals from receptors at the plasma membrane. Recent reports point to a novel localization of G proteins at the mitochondria and other endomembranes where they regulate the physiology of these organelles. In particular, the Gαq subfamily is required to keep the proper balance between mitochondria fusion and fission acting at both outer and inner membrane, among other functions. In order to unveil the putative effectors of Gαq that mediate those effects at the mitochondria, our group has undertaken a mass-spectrometry analysis based on Gαq immunoprecipitates from cellular endomembranes. The “mito- interactome” study provided evidence of Gαq interaction with the armadillo domain-containing proteins Alex3 and Armc10. Subsequent immunoprecipitation and pull-down studies demonstrated a specific interaction of Gαq with the mitochondrial Rho GTPase 1 (Miro1) and both milton adaptor proteins TRAK1 and 2, that couple mitochondria to kinesin and dynein motor proteins and constitute the main regulators of mitochondrial transport in neurons. To analyze the physiological role of those interactions, we have performed tracking analysis of mitochondria along the axons of hippocampal neurons overexpressing Gαq or its constitutive-active mutant, GαqR183C, as well as activating a Gαq-specific GPCR (DREADD) with its specific agonist. The results of these studies reveal a significant increase in anterograde movement upon Gαq expression, whereas Gαq activation by either expressing the active-mutant or activating the Gαq-specific GPCR induces mitochondrial arrest. In contrast, depletion of Gαq using short-hairpin RNAs increases the number of motile mitochondria and their speed and promotes retrograde transport. Both activation of Gαq or its depletion alter mitochondrial dynamics including fusion/fission events, whereas expression of active-Gαq also alters neuronal physiology by reducing their complexity and dendritic branching. In summary, our group postulates a new non-canonical mitochondrial function of Gαq acting as a molecular switch through its association with Alex3, Miro1 and TRAK2. Gαq would associate to Alex3 and Miro1 to allow mitochondrial movement, whereas its GTP-bound conformation would associate to TRAK2 to halt motility. This process would be regulated by Alex3, which could play crucial roles as an adaptor for the protein complex and Gαq transactivation.

Les adénovirus ont une double nature, soit comme pathogène omniprésent qui peuvent occasionnellement causer des maladies soit comme vecteurs utilisés de transfert de gène. À nos connaissances, les 30 premières minutes depuis la liaison au récepteur jusqu'à l'arrivée au pore nucléaire sont identiques pour le pathogène comme pour le vecteur. L'objectif de ma thèse était de comprendre les mécanismes impliqués dans la liaison au récepteur, l'internalisation, l'échappement et le trafic endosomal vers le MTOC. J'ai d'abord étudié le mécanisme impliqué dans l'hémagglutination des virus à tropisme pour CAR et à tropisme pour SA. J'ai identifié la présence de CAR sur les érythrocytes humains et montré qu'il était le principal responsable de l'agglutination induite par les virus à tropisme pour CAR. De plus, j'ai montré que la présence de CAR sur les érythrocytes pouvait piéger le virus dans le sang et ainsi empêcher l'infection au niveau du foie. Dans un deuxième temps, j'ai participé à la caractérisation du rôle de la protéine VI et la translocation du virus au MTOC. Nous avons montré que Nedd4 était impliqué dans le ciblage du virus au MTOC via l'ubiquitination de la protéine VI. Enfin, j'ai travaillé sur le neurotropisme de CAV-2 et caractérisé sa localisa tion subcellulaire au niveau des synapses. J'ai montré qu'une partie de CAR était localisée dans des radeaux lipidiques à la synapse et que CAV-2 entrait via la voie de recyclage des vésicules synaptiques Adenoviruses have a dual nature as ubiquitous pathogens that occasionally cause life-threatening disease and their use as gene transfer vectors. To the best of our current knowledge, the first 30 min from binding to nuclear pore docking of both wild-type virus and vector are identical. The goal of my thesis is to understand different mechanisms involved in receptor binding, internalization, endosomal escape and trafficking to the MTOC. First I studied the mechanism involved in hemagglutination of CAR-tropic and SA-tropic viruses. I identified the presence of CAR on human erythrocytes and showed that it was the main responsible for the agglutination mediated by CAR-tropic viruses. Moreover, I show that CAR on erythrocytes can sequester virus in the bloodstream and block liver infection. In a second part I participated to the characterization of the role of the protein VI and the translocation of HAd to the MTOC. We showed that Nedd4 was involved in the targeting of the virus to MTOC through ubiquitination of this protein VI. Finally, I worked on the neurotropism of CAV-2 and characterize its subcellular localization at the synapse. I showed that a part of CAR was localized in lipid raft at the synapse and enter through the synaptic vesicle-recycling pathway

Abstract:

La croissance neuritique est un mécanisme complexe qui fait toujours l'objet d'intenses investigations. Les donnés actuelles ont permis de mettre en évidence l'implication de trois mécanismes principaux dans la croissance neuritique : i) la dynamique du cytosquelette, ii) le trafic intracellulaire et l'apport membranaire au niveau du cône de croissance et iii) la signalisation cellulaire, principalement via la voie MAPK-ERK1/2, qui abouti à la régulation de la transcription.La PLD1 et son produit l'acide phosphatidique semblent être au centre de voies majeures impliquées dans le développement neuronal. Mes travaux ont permis d'approfondir nos connaissances sur le rôle cellulaire de la PLD1 au cours de la croissance neuritique. J'ai montré que la PLD1 en collaboration avec la kinase RSK2 régule la fusion des vésicules positives pour Ti-VAMP/VAMP7 au cours de la croissance neuritique. D'autre part, j'ai établi que la PLD1 joue un rôle important dans le maintien de la signalisation endosomale de la voie MAPK-ERK1/2-RSK2-CREB induite par les neurotrophines. J'ai également montré que la PLD1 régule l'activation de mTOR/p70S6K en réponse au BDNF. La dérégulation des voies MAPK-ERK1/2 et mTOR/p70-S6K pourraient être à la base de la réduction de l'arborisation dendritique et de la maturation des épines dendritique observée dans les neurones corticaux Pld1-/- en culture. En plus de l'implication de RSK2 dans la régulation de la PLD1, j'ai également montré que la PLD1 régule l'activation de RSK2 en réponse aux neurotrophines, probablement via une boucle de rétrocontrôle. Ainsi les donnés obtenus suggèrent un lien fort entre les deux protéines au cours du développement neuronal. A la lumière de ces donnés, un dysfonctionnement de ce mécanisme pourrait expliquer le retard mental observé chez les patients atteints du syndrome de Coffin-Lowry causé par la perte de l'activité kinase de RSK2. D'autre part, les résultats obtenus suggerent un rôle de la PLD1 dans l'exocytose des vésicules.

Abstract:

Nach dem Eintritt in den Wirtsorganismus und initialer Replikation infizieren Alphaherpesviren Neuronen zur weiteren Ausbreitung im Nervensystem und zur Etablierung einer Latenz. Dazu werden die Viruspartikel innerhalb der Axone retrograd von der Peripherie zum neuronalen Zellkörper transportiert. Die umgekehrte Richtung beschreibt den Weg des anterograden Transports vom Zellkörper zur Synapse für weitere Infektionen von Neuronen höherer Ordnung oder zurück zur Peripherie. Der retrograde intraaxonale Transport ist gut untersucht. Dagegen wird über den anterograden Transport kontrovers diskutiert. Zwei verschiedene Transportmodelle werden vermutet. Das „Married Model“ postuliert, dass umhüllte Virionen innerhalb von Vesikeln entlang des Axons transportiert werden. Die Freisetzung der Partikel erfolgt an der jeweiligen Synapse durch Endocytose. Das „Subassembly Model“ geht dagegen davon aus, dass einzelne Virusstrukurkomponenten (Nukleokapsid, Hülle) entlang des Axons transportiert werden. Der Zusammenbau und die Freisetzung erfolgt am Axonterminus bzw. an der Synapse (in vivo) oder am Wachstumskegel (in vitro) oder an speziellen Auftreibungen des Axons, den sogenannten Varicosities. Nach Infektion eines neuronalen Explantatsystems mit dem Pseudorabiesvirus (PrV) konnten ultrastrukturell umhüllte Virionen in Vesikeln detektiert werden und so der Nachweis der Gültigkeit des „Married Model“ als vorherrschendes Transportmodell geführt werden. Dagegen ist die Situation beim prototypischen Alphaherpesvirus, dem Herpes Simplex Virus Typ 1 (HSV-1), weiterhin ungeklärt. Aufgrund der zahlreichen unterschiedlichen Analysemethoden und -systeme war ein direkter Vergleich der beiden Viren bislang nicht möglich. Daher sollte in dieser Arbeit ein standardisiertes neuronales Kultursystem genutzt werden, um vier verschiedene HSV-1 Stämme im Vergleich zu PrV zu untersuchen. Für die Infektionen wurden sowohl Neuronen aus dem oberen Cervikalganglion als auch aus Spinalganglien genutzt. So konnte gezeigt werden, dass in Neuronen, welche mit den HSV-1 Stämmen HFEM, 17+ und SC16 infiziert waren ca. 75% als umhüllte Virionen in Vesikeln und ca. 25% als nackte Kapside vorlagen. Ingesamt war die Anzahl der Viruspartikel in HSV-1 infizierten Neuronen signifikant geringer als in PrV infizierten Kulturen. Überraschenderweise zeigten mit HSV-1 KOS infizierte Neuronen ein reverses Bild. Hier lagen nur 25% der Viruspartikel als umhüllte Virionen in Vesikeln vor, während 75% als nackte Kapside detektiert wurden. Dieser unerwartete Phänotyp sollte auf molekularbiologischer Ebene genauer untersucht werden. Dabei wurde auf die Genregion von US9 fokussiert. Das von US9 codierte Membranprotein spielt eine wichtige Rolle während des Zusammenbaus der Virionen und bei anschließenden axonalen anterograden Transportvorgängen. In dieser Arbeit konnte gezeigt werden, dass das HSV-1 KOS Genom durch verschiedene Basenaustausche an der vorhergesagten TATA-Box von US9 eine Mutation aufweist. Zusätzlich trägt das offene Leseraster durch eine weitere Mutation ein vorzeitiges Stopcodon auf und wird dadurch auf 58 Kodons reduziert, im Gegensatz zu anderen HSV-1 Stämmen, wo es 91 Kodons umfasst. Die Mutation an der TATA-Box verändert auch das ursprüngliche Stopcodon vom US8a Gen, was zur einer Verlängerung von ursprünglich 161 zu 191 Kodons führt. In Northern Blot Analysen konnte eine reduzierte Transkription von US9 in HSV-1 KOS infizierten Zellen detektiert werden. In HSV-1 KOS infizierten Zellen konnten mittels eines spezifischen Antiserums gegen US9 im Western Blot kein Genprodukt nachgewiesen werden. Auch Immunfluoreszenzanalysen zeigten, dass das abgeleitete verkürzte Protein offenbar nicht stabil exprimiert wird. Dagegen konnten Western Blot Analysen die Vergrößerung des pUS8a bestätigen. Der beobachtete auffällige intraaxonale Phänotyp könnte somit durch die Mutation des US9 Protein erklärt werden. Zusammenfassend wurde in dieser Arbeit gezeigt, dass auch bei HSV-1 vorwiegend das „Married Model“ für den anterograden intraaxonalen Transportweg bevorzugt wird und somit beide Alphaherpesviren, HSV-1 und PrV, denselben Transportweg nutzen.

Abstract:

Au cours de cette thèse, nous avons développé des méthodes d’IRM à très haut champ (7T) permettant de caractériser les modifications morphologiques et fonctionnelles survenant chez différents modèles animaux de la maladie d’Alzheimer. Chez le microcèbe murin, un primate non-humain de très petite taille, nous avons étudié le vieillissement cérébral par micro-IRM. Les animaux âgés présentent une réorganisation des sous-régions hippocampiques, associant une croissance du gyrus denté et une atrophie de la corne d’Ammon, même en l’absence d’atrophie hippocampique globale détectable. Des hypointensités focales T2* apparaissent progressivement dans le cerveau des animaux âgés, correspondant le plus souvent à des microhémorragies, et parfois à des dépôts amyloïdes diffus. Chez deux modèles murins transgéniques d’amyloidogenèse (lignée 5XFAD) et de tauopathie (lignée JNPL3[P301L]), nous avons développé une méthode d’estimation non invasive du transport neuronal par IRM injectée au Manganèse (Manganese-Enhanced MRI ou MEMRI). Les souris JNPL3[P301L] présentent une altération progressive du transport neuronal, qui est corrélée de façon significative avec le degré de tauopathie en histologie. Chez les souris 5XFAD, de façon surprenante, le transport neuronal semble préservé, alors qu’il décline avec l’âge chez les souris contrôles. Cette apparente préservation du transport neuronal chez les souris 5XFAD pourrait correspondre à des phénomènes d’excitotoxicité liés à l’amyloïdogenèse

Synaptic plasticity modulates development and strengthening of new neuronal connections, or synapses, upon learning and is based on local protein synthesis next to activated neuronal spines. Thousands of mRNA species are transported from the soma of neuron to dendrites, in order to be translated on demand. The mechanisms of this mRNA transport remain poorly understood. Mutations of Fragile X-mental retardation protein (FMRP) cause a spectrum of mental retardation disorders. Among other functions, FMRP mediates signal-mediated mRNA transport and local translation in dendrites. In multiple attempts to understand how FMRP is implicated in mRNA transport, there were identified few motor protein candidates. Besides controversy in the literature, none of these motor proteins was demonstrated to bind directly and transport FMRP. In my PhD studies, I employed in vitro reconstitution assays, coupled to Total Internal Reflection (TIRF) microscopy, to test, which of the proposed candidate motors can transport FMRP along the microtubules and whether FMRP can co-transport mRNA molecules. In order to understand the biochemistry of FMRP-mediated mRNA transport, I have purified and tested motor proteins from three Kinesin subfamilies. In this PhD thesis, I am reporting that FMRP binds directly to and is transported by Kinesin-2 motor (KIF3A/C heterodimer), but not by the other tested motors. Mutational analysis of FMRP suggests that its C-terminal region plays the biggest role in Kinesin-2 binding, and that this interaction does not depend on the RGG box region, known to recognise the G-quadruplex structure of FMRP’s mRNA targets. These results suggest that mRNA and motor binding by FMRP are not mutually exclusive, and thus FMRP must be capable to mediate mRNA transport. I also show that KIF3A/C motor binds several mRNA targets, with and without G-quadruplex structure, and that G-quadruplex mRNA competes with FMRP for motor binding. These results raise many questions that I address in the Discussion part of the thesis. This work is the first of its kind, to my knowledge, to systematically test kinesin motor proteins for direct interaction with FMRP and to reconstruct an FMRP transport complex. I conclude that FMRP binds directly to the Kinesin-2 motor and that this complex moves processively along the microtubules. This complex is still missing its cargo, mRNA, which will be investigated beyond the scope of this PhD thesis. I analyse the speeds of used kinesin motors and compare them to the literature. In the end, I discuss possible reasons why FMRP was not binding the G-quadruplex mRNA in my experimental conditions and outline the caveats of in vitro reconstitution assays. La plasticidad sináptica modula el desarrollo y el fortalecimiento de nuevas conexiones neuronales, o sinapsis, al aprender y se basa en la síntesis local de proteínas al lado de las espinas neuronales activadas. Miles de especies de ARNm son transportadas desde el soma de la neurona a las dendritas, para ser traducidas bajo demanda. Los mecanismos de este transporte de ARNm siguen siendo poco conocidos. Las mutaciones de la proteína Fragile X-mental Retardation (FMRP) causan un espectro de trastornos de retraso mental. Entre otras funciones, FMRP media el transporte de ARNm inducido por señal y la traducción local en las dendritas. En múltiples intentos de comprender cómo FMRP está implicada en el transporte de ARNm, se identificaron algunos candidatos de proteínas motoras. Además de la controversia en la bibliografía, no se demostró que ninguna de estas proteínas se uniera directamente y transportara FMRP. En mis estudios de doctorado, he utilizado ensayos de reconstitución in vitro, junto con la microscopía de reflexión interna total (TIRF), para probar cuál de los motores candidatos propuestos puede transportar FMRP a lo largo de los microtúbulos y si FMRP puede co-transportar moléculas de ARNm. Para comprender la bioquímica del transporte de ARNm mediado por FMRP, he purificado y probado proteínas motoras de tres subfamilias de kinesina. En esta tesis doctoral, presento que FMRP se une directamente y es transportada por el motor Kinesin-2 (heterodímero KIF3A/C), pero no por los otros motores probados. El análisis de mutaciones de FMRP sugiere que su región C-terminal juega el papel más importante en la unión con Kinesin-2, y que esta interacción no depende de la región de RGG box, conocida por identificar la estructura de G.quadruplexde de los ARNm diana de FMRP. Estos resultados sugieren que FMRP es capaz de unirse al motor y transportar ARNm simultáneamente. También muestro una observación de que el motor KIF3A/C se une a varios ARNms, con y sin estructura de G-quadruplex, y que el ARNm con G-quadruplex compite con FMRP por la interacción con la proteína motora. Estos resultados plantean muchas preguntas que abordo en la parte de Discusión de esta tesis. Este trabajo es el primero de su tipo, en mi conocimiento, para probar sistemáticamente la interaccion directa de FMRP con las proteínas motoras de kinesina y para reconstruir un complejo de transporte de FMRP. Concluyo que FMRP se une directamente al motor Kinesin-2 y que este complejo se mueve procesivamente a lo largo de los microtúbulos. A este complejo todavía le falta su carga, ARNm, que se investigará más allá del alcance de esta tesis doctoral. Analizo las velocidades de los motores de kinesina utilizados y los comparo con la bibliografía. Al final, discuto las posibles razones de por qué FMRP no se estaba uniendo el ARNm de G-quadruplex en mis condiciones experimentales y describo los escollos en utiliza de los ensayos de reconstitución in vitro.

Abstract:

The synapse, the minimal element required for interneuronal communication in the nervous sytems, is a structure with a great deal of plasticity, capable of undergoing changes that alter transmission strength, and even forming new connections. This property has great implications for a number of processes, including circuit formation and learning and memory. However, the proteins behind this synaptic plasticity are still not fully understood. To uncover and characterize the proteins that regulate the plastic nature of the synapse, I turned to the Drosophilalarval neuromuscular junction (NMJ), a powerful and accessible model system. I began by examining synaptic cell adhesion, as Cell Adhesion Molecules (CAMs) have long been implicated in synaptic outgrowth as well as learning and memory. CAMs have traditionally been thought of as molecules that mediate cell adhesion between the pre- and postsynaptic membrane. However, through the course of the studies presented here I demonstrate a CAM function that goes beyond simple cell adhesion, acting as a receptor that transduces adhesive signals to the intracellular space. In particular, I have demonstrated a role for the Drosophila CAM, Fasciclin II(FasII), in a signaling complex involving the Amyloid Precursor Protein-Like (APPL) and the Drosophila homolog of X11/MINT/Lin-10 (dX11). Further results show that deletion of either APPL or dX11 inhibits the FasII mediated outgrowth. These studies show that during NMJ expansion the transinteraction between FasII molecules in the pre- and postsynaptic membrane results in the recruitment of APPL and dX11 to the presynaptic cell surface, and the initiation of a signaling cascade that leads to bouton outgrowth. The next question addressed here was regarding the cytoskeletal changes that must occur during synapse remodeling. In particular I centered on the evolutionarily conserved cell polarity complex aPKC-Par3-Par6, which is know to regulate axon growth, the cell cytoskeleton during polarized cell division, and learning and memory. To understand the role of the cytoskeleton during NMJ expansion, I examined the organization of microtubules and actin during this process. Further, I identified atypical protein kinase C (aPKC) as a regulator of microtubule dynamics. I found that aPKC is required for regulating the degree of stabilization of synaptic microtubules. This stabilization requires the Microtubule Associated Protein-1B (MAP1B) homolog Futsch, which I demonstrated was required for aPKC to associate with and stabilize the microtubule cytoskeleton. The process of synaptic expansion not only requires modifications to the presynapse, but to the postsynapse as well. Previous work demonstrates that levels of the scaffolding proteins DrosophilaMembrane Associated Guanlyate Kinase (MAGUK) protein Discs-large (DLG), as well as the vertebrate homolog Postsynaptic Density-95 (PSD-95), which are concentrated at synapses, determine the size of postsynaptic membranes. To identify the underlying mechanisms of the regulation of postsynaptic size, we performed a yeast two hybrid screen, searching for DLG interacting proteins. We found a novel interaction between DLG, and a t-SNARE, GUK-interacting Syntaxin (Gtaxin; GTX), and went on to demonstrate that this interaction is required for proper postsynaptic membrane addition. Strong hypomorphic mutations in either dlg or gtx show a dramatic reduction in postsynaptic expansion. Overexpression of DLG produces an increase of synaptic GTX, as well as an increase in postsynaptic size, and an increased formation of GTX positive SNARE complexes. Taken together, these observations suggest that the MAGUK DLG regulates postsynaptic membrane addition by modulating the formation of a SNARE complex of the t-SNARE Gtaxin, and by targeting GTX to sites of postsynaptic membrane addition. In summary, the studies performed in this thesis probe a trans-synaptic adhesion based signaling complex required for presynaptic expansion, a specific pathway for dynamic microtubule stabilization required for pre- and postsynaptic expansion, and how a scaffolding protein regulates postsynaptic membrane expansion. These processes are all interconnected to maintain the efficacy of the synapse. The studies conducted revealed important information about how these processes are accomplished, and constitute an important step to elucidate the mechanisms by which synapse plasticity occurs at the level of single synaptic terminals.

Abstract:

Glutamate transporters have a homotrimeric subunit structure with a large central water-filled cavity that extends partially into the plane of the lipid bilayer (Yernool et al., 2004). In addition to uptake of glutamate, the transporters also mediate a chloride conductance that is gated by Na+ and glutamate. Our data indicate that glutamate binding sites, transport pathways, and chloride channels reside in individual subunits in the trimer and function independently and that the anion channel is gated by alkali cations binding from either side of the membrane. We also investigated conformational changes during glutamate binding by incorporating a fluorescent probe into a site near the postulated external gate (HP2) of a mutant transporter that can bind but not transport L-Glu. Fluorescence changes were observed upon ligand binding that strongly depended on the number of subunits labeled; this suggests quenched fluorophore dimers form at the center of the trimer that are subject to HP2 loop closure upon substrate binding. This supports a model of gate motion that is also consistent with recent x-ray structural data (Boudker et al., 2007). Finally, we propose that the large aqueous central cavity in the trimeric complex (Yernool et al., 2004) may function to restrict the diffusion of molecules near the three ligand binding sites, resulting in an increase in the probability of rebinding.

L’identification de biomarqueurs des maladies mentales telles que l’autisme, la schizophrénie ou la maladie d’Alzheimer, est d’une importance capitale non seulement pour établir un diagnostic objectif, mais aussi pour suivre l’effet des traitements. La création et le maintien de fonctions neuronales sub-cellulaires, telle que la plasticité synaptique, sont fortement dépendants du transport intraneuronal, essentiel pour acheminer d’importants composants à des positions spécifiques. Un transport actif défaillant semble être partiellement responsable d’anomalies de la plasticité synaptique et de la morphologie neuronale présentes dans de nombreuses maladies neuropsychiatriques. Cette thèse décrit (i) la mise au point d’une méthode de quantification du transport intraneuronal reposant sur le suivi de nanoparticules de diamants fluorescents (fNDs); (ii) l’application de cette technique simple et faiblement invasive à l’analyse fonctionnelle de variants génétiques associés à des maladies neuropsychiatriques. Ce manuscrit comporte quatre chapitres. Le premier détaille l’architecture polygénique complexe des maladies mentales et démontre la pertinence d’étudier le transport intraneuronal. Les deuxième et troisième chapitres sont dédiés à la méthode et détaillent les stratégies d’internalisation des fNDs, les outils de quantification du transport intraneuronal et la validation de la technique. La forte brillance, la photo-stabilité parfaite et l’absence de toxicité cellulaire font des fNDs un outil de choix pour étudier la dynamique du transport intraneuronal sur une durée d’observation de plusieurs heures avec une haute résolution spatiotemporelle et une bonne puissance statistique. Enfin, dans le quatrième chapitre, nous appliquons cette nouvelle méthode d’analyse fonctionnelle pour étudier l’effet de variants génétiques associés à l’autisme et à la schizophrénie. Pour cela, nous utilisons des lignées de souris transgéniques ayant une faible surexpression des gènes MARK1 et SLC25A12, ainsi que des AAV-shRNA pour induire une haplo-insuffisance du gène AUTS2. Abnormalities in such active transport are thought to be partly responsible for synaptic plasticity and neuronal morphology impairment found in many neuropsychiatric and neurodegenerative diseases. This thesis reports (i) the development of a quantification technic of intraneuronal transport based on fluorescent nanodiamonds (fNDs) tracking; (ii) the application of this simple and minimally invasive approach to the functional analysis of neuropsychiatric disease-related genetic variants.This manuscript falls into four chapters. The first one details the complex polygenic architecture of mental disorders and demonstrates the disease relevance of monitoring the intraneuronal transport. The second and the third chapters are dedicated to the nanodiamond-tracking assay and describe the fNDs internalisation strategies, the spatiotemporal quantitative readouts and the validation of the technic. The high brightness, the perfect photostability and the absence of cytotoxicity make fNDs a tool of choice to perform high throughput long-term bioimaging at high spatiotemporal resolution. Finally, in the fourth chapter, we apply this new functional analysis method to study the effect of genetic variants associated to autism and schizophrenia. We established transgenic mouse lines in which MARK1 and SLC25A12 genes were slightly overexpressed, and AAV-shRNA to induce AUTS2 gene haploinsufficiency. Our molecular diagnosis assay proves sufficiently sensitive to detect fine changes in intraneuronal transport dynamic, paving the way for future development in translational nanomedicine

Abstract:

Dynamic mitochondrial transport into axons and dendrites of neuronal cells is critical for sustaining neuronal excitability, synaptic transmission, and cell survival. Failure of mitochondrial transport is the direct cause of some neurodegenerative diseases, and an aggravating factor for many others. Mitochondrial transport regulation involves many proteins; factoring prominently among them are the atypical mitochondrial GTPase Miro and the Milton/TRAK adaptor proteins, which link microtubule (MT) motors to mitochondria. Motors of the kinesin family mediate mitochondrial transport towards the plus ends of microtubules, while motors of the dynein family mediate mitochondrial transport towards the minus ends. Selective use of these motors determines the ultimate subcellular distribution of mitochondria, but the underlying control mechanisms remain poorly understood. Drosophila Miro (dMiro) is required for kinesin-driven transport of mitochondria, but its role in dynein-driven transport remains controversial. In Chapter 2 of this study, I show that dMiro is also required for the dynein-driven transport of mitochondria. In addition, we used the loss-of-function mutations dMiroT25N and dMiroT460N to analyze the function of dMiro's N- and C-terminal GTPase domains, respectively. We show that dMiroT25N causes lethality and impairs mitochondrial distribution and transport in a manner indistinguishable from dmiro null mutants. Our analysis suggests that both kinesin- and dynein-driven mitochondrial transport require the activity of Miro's N-terminal GTPase domain, which likely controls the transition from a stationary to a motile state irrespective of the transport direction. dMiroT460N reduced only dynein motility during retrograde axonal transport but had no effect on distribution of mitochondria in neurons, indicating that the C-terminal GTPase domain of Miro most likely has only a small modulatory role on transport. Furthermore, we show that commonly used substitutions in Miro's GTPase domains, based on the constitutively active Ras-G12V mutation, appear to cause neomorphic phenotypic effects which are probably unrelated to the normal function of the protein. In mammalian neurons, kinesin and dynein motors are linked to mitochondria via a Miro complex with the adapter proteins TRAK1 and TRAK2, respectively. Differential linkage of TRAK-motor complexes provides a mechanism for determining the direction of transport and controlling mitochondrial distributions within the cell. Drosophila has only one TRAK gene homolog, Milton, which expresses several protein isoform. Milton has been previously been shown to facilitate mitochondrial transport by binding to kinesin and dMiro, a role analogous to TRAK1. However, the question whether Milton might be able mediate dynein-based transport in a manner similar to TRAK2 has remained unknown. In Chapter 3 of this study, I show that protein isoforms A and B of Milton, generated through alternative mRNA splicing, facilitate differential motor activities analogous to mammalian TRAKs. Specifically, overexpression (OE) of Milton-A caused a mitochondrial redistribution and accumulation at axon terminals, which requires kinesin-driven MT plus end directed transport; while OE of Milton-B caused a redistribution of axonal mitochondria into the soma, which requires dynein-driven MT minus end directed transport. I further show that Milton-motor complex binding to mitochondria requires Miro exclusively, and that transport with either of the motor complexes absolutely requires the activity of Miro's N-terminal GTPase domain. Together, these results suggest that Miro controls the transition of mitochondria from a stationary to a motile phase. Thereafter the direction of transport is likely determined by an alternative binding of opposing Milton/TRAK-motor complexes to Miro, a process which appears to be regulated by a Miro-independent mechanism.

Le transport intracellulaire implique le mouvement de diverses vésicules et organites par des moteurs moléculaires le long des microtubules, et est donc un processus crucial dans toutes les cellules eucaryotes. Les cellules hautement différenciées, complexes et à longue durée de vie telles que les neurones posent un défi particulier pour le maintien d'un transport efficace et étroitement contrôlé tout au long de la vie d'un organisme. Les défauts du transport axonal ont été impliqués dans de nombreuses maladies neurodégénératives et sont actuellement considérés comme l'un des premiers événements de la voie pathogène. Les mécanismes de contrôle du transport intra-neuronal pourraient donc jouer un rôle important dans l'homéostasie neuronale. Un mécanisme qui a été proposé pendant de nombreuses années pour contrôler le transport intracellulaire sont les modifications post-traductionnelles des voies de transport - les microtubules. Dans ma thèse de doctorat, j'ai testé l'hypothèse qu'une modification de tubuline particulièrement enrichie dans les neurones, la polyglutamylation, contrôle le transport axonal.Au cours de mon doctorat, j'ai établi toutes les méthodes nécessaires pour obtenir des neurones avec des niveaux différents de polyglutamylation en utilisant plusieurs modèles de souris. J'ai ensuite mesuré le transport de différentes cargaisons dans ces neurones. Premièrement, j'ai mesuré le transport des mitochondries et démontré qu'une polyglutamylation accrue de la tubuline, qui cause la dégénérescence des neurones, réduit la motilité des mitochondries. Inversement, dans les neurones à polyglutamylation diminuée, le transport des mitochondries était augmenté. Ces résultats suggèrent que la polyglutamylation de la tubuline est un régulateur dynamique du transport des mitochondries. Pour étudier la spécificité de la régulation du transport par la polyglutamylation, j'ai ensuite mesuré le transport d'autres cargos axonaux. Le transport des lysosomes, des endosomes tardifs et des vésicules de BDNF a été, comme pour les mitochondries, affecté négativement par l'hyperglutamylation.En conclusion, j'ai démontré que la polyglutamylation de la tubuline pourrait être un mécanisme général contrôlant le transport axonal. Considérant nos résultats antérieurs, qui montrent que l'hyperglutamylation induit la neurodégénérescence, les défauts du transport axonal pourraient être l'un des principaux mécanismes moléculaires qui induisent la dégénérescence des neurones avec les microtubules hyperglutamylés Intracellular transport involves transporting various vesicles and organelles by molecular motors along the microtubules, and is thus a crucial process in all eukaryotic cells. Highly differentiated, complex and long-lived cells such as neurons pose a particular challenge for the maintenance of an efficient, tightly controlled transport. Defects in axonal transport have been implicated in many neurodegenerative disorders, and are currently considered as one of the early events in the pathogenic pathway. Mechanisms controlling intra-neuronal transport could thus be important players in neuronal homeostasis. A mechanism that was for many years proposed to control intracellular transport is the posttranslational modification of microtubule tracks. In my PhD thesis I tested the hypothesis that one tubulin modification that is particularly enriched in neurons, polyglutamylation, controls axonal transport.During my PhD, I have established all the necessary methods to obtain neurons with differential levels of polyglutamylation using different mouse models. I have then measured the transport of different cargoes in these neurons. First, I have measured mitochondria transport, and demonstrated that increased tubulin polyglutamylation, as found in neurons that degenerate, reduces the overall motility of mitochondria. Inversely, in neurons with decreased polyglutamylation, I have shown that mitochondria transport is increased. These results suggest that tubulin polyglutamylation is a dynamic regulator of mitochondrial transport. To investigate the specificity of transport regulation by tubulin polyglutamylation, I next measured the transport of other axonal cargoes. Transport of lysosomes, late endosomes and BDNF vesicles was, similar to mitochondria, negatively affected by hyperglutamylation.In conclusion, I have demonstrated that tubulin polyglutamylation could be a general tuning mechanism for axonal transport. Considering our earlier findings, which show that hyperglutamylation induces neurodegeneration, defects in axonal transport could be one of the key molecular mechanisms that induce degeneration of neurons with hyperglutamylation

Les etudes pharmacologiques, sur membranes striatales et sur neurones en cultures primaires, ont montre que la btcp, derive de la phencyclidine (pcp), se lie avec une haute affinite sur le complexe de recapture de la dopamine, dont elle inhibe selectivement le transport. L'utilisation de la btcp, comme outil de marquage des sites de recapture de la dopamine, a permis de localiser ces derniers dans les zones de l'encephale de rat adulte riches en elements dopaminergiques (striatum et substance noire) et sur l'ensemble du neurone exprimant la dopamine en culture (au niveau des membranes terminales, neuritiques et somatiques). Un traitement chronique, par la btcp, entraine des modifications metaboliques (reduction de la synthese et accumulation accrue de la dopamine), qui influencent le developpement des neurones dopaminergiques in vitro. Ces effets sont modules par la presence de cellules striatales cibles dans la culture, qui previent les effets de la drogue. Abstract:

La prévision de trajectoires d'avions est un problème crucial pour les systèmes de gestion du traffic aérien. Des méthodes de prévision utilisant des modélisations d'avion existent déjà mais nécessitent beaucoup de paramètres en n'offrant qu'une précision moyenne. C'est pourquoi nous développons une nouvelle approche nútilisant que peu d'informations initiales. La méthode choisie est basée sur l'utilisation de réseaux de neurones auxquels on fait apprendre un ensemble de trajectoires avant de les utiliser pour en prédire de nouvelles. Nous développons trois méthodes différentes qui permettent une prédiction à long terme dans le plan vertical et à court et moyen terme dans le plan horizontal. L'une d'elles est capable d'intégrer de nouvelles données au fur et à mesure que l'avion vole, ce qui lui permet de réagir aux éventuels changements de trajectoire et ainsi de parfaire la prévision. Les deux autres réalisent la prédiction même lorsque l'avion ne vole pas. Nous introduisons aussi une structure de type mélange hiérarchique d'experts qui permet de regrouper le savoir emmagaziné dans plusieurs réseaux, ainsi, il est possible de se passer de la connaissance du type d'avion étudié. Les méthodes utilisant des réseaux de neurones sont ensuite comparées aux méthodes à base de modèles avion. Nous montrons alors que les réseaux de neurones donnent de bien meilleurs résultats tout en permettant une prédiction très rapide. De plus, ne nécessitant que très peu de paramètres au départ, ils sont applicables sur un plus grand nombre de cas.
