A novel series of 1,3,5-trisubstituted pyrazole derivatives were synthesized by the cycloaddition reaction between the electron rich N-substituted aryl hydrazones and nitro-olefins. Different N-substituted aryl hydrazones (1a-j) on reacting with 4-methyl-β-nitrostyrene (2) in ethylene glycol or trifluoroethanol-trifluoro acetic acid mixture (for acid stable hydrazones) at 120 ºC, yielded various 1,3,5-trisubstituted pyrazoles (3a-j). Analytical method was developed for the estimation of Sofosbuvir and Daclatasvir drug substance by liquid chromatography. The chromatographic separation was achieved on C18 column (XTerra RP18 150*4.6, 5um) at ambient temperature .The separation achieved employing a mobile phase consists of 0.1%v/v Trifluoro acetic acid in water: Acetonitrile (60:40). The flow rate was 1.0 ml/ minute and ultra violet detector at 275nm. The average retention time for Sofosbuvir and Daclatasvir found to be 2.09 and 3.50 min. The synthesis of previously unknown unsaturated aromatic oxyethers was carried out by reaction of chloromethylpropargyl(allyl) ether with phenacyl bromide, with the participation of the chiral catalyst - (+)-benzotetramisole, and their configurations were also established.
