Gene-gene or gene-drug interactions are typically quantified using fitness as a readout because the data is continuous and easily measured in high-throughput. However, to what extent fitness captures the range of other phenotypes that show synergistic effects is usually unknown. Using Saccharomyces cerevisiae, and focusing on a matrix of DNA repair mutants and genotoxic drugs, I quantified 76 gene-drug interactions based on both mutation rate and fitness and find that these parameters are not connected. C. elegans worms with mutations in the gene CLK-1 develop slowly and have an extended lifespan. CLK-1 encodes a mitochondrial protein that is responsible for the hydroxylation of 5-demethoxyubiquinone (DMQ), the penultimate step of ubiquinone (Coenzyme-Q or UQ) biosynthesis. Structural homologues of CLK-1 are found in mammals, fruit flies, yeast and some types of bacteria. In principle, a pre-constructed library of all possible short oligonucleotides could be used to construct many distinct gene sequences. This approach requires computational methods to accurately determine the assembly procedure, but relieves the current technological constraints of custom oligonucleotide synthesis. The present study investigated whether this increase in amylase activity is a result of increased pancreatic amylase messenger RNA (mRNA) levels. Currently there has been insufficient study of the fundamental understanding of these motifs which has resulted in a lack of consensus of their role and presence in biology.
