Abstract:

Human herpesvirus type 7 (HHV-7), with its high prevalence and apparently non pathogenic nature, is believed to be a virus very well adapted to humans. It is therefore useful to study the transcription patterns of the virus and the host to understand the life cycle and pathogenesis (or the lack of it) of HHV-7 infection. In order to obtain the global pattern of host and viral gene expression, we have made a DNA microarray containing probes that target all 86 predicted HHV-7 open reading frames and approximately 1000 cellular genes. This thesis describes the isolation and characterization of the U24 membrane protein encoded by Human Herpesvirus type-6 (HHV-6), obtained from an E. coli recombinant expression system. HHV-6 infection has been previously associated with the disease multiple sclerosis (MS), and the U24 protein is of interest because it has a seven amino-acid sequence (PRTPPPS) identical to myelin basic protein (MyBP), a candidate auto-antigen in MS. A infecção primária com o herpesvirus humano tipo 6 (HHV-6) pode resultar no exantema súbito, encefalite e recorrentes complicações. Pesquisas demonstram que crianças obtêm os anticorpos contra o HHV-6 antes dos dois anos de idade. Após a infecção primária, o DNA do HHV-6 permanece latente em linfócitos, saliva ou líquor. Spinocerebellar ataxia type 7 (SCA7) is a hereditary neurodegenerative disorder affecting the cerebellum, pons and retina. SCA7 patients present with gait ataxia and visual impairment as the main symptoms. Human herpes virus type 8 (HHV-8) is strongly implicated as the etiological agent of Kaposi’s sarcoma (KS). The incidence of KS in South Africa is increasing in parallel with the HIV-1 epidemic. Molecular and serological prevalence of HHV-8 in HIV-1 infected individuals with and without KS was investigated. DNA fragments from ORF26 (capsid, 330BAM233) and ORF75 (tegument) regions were used to determine the prevalence of HHV-8 DNA in peripheral blood mononuclear cells (PBMC) from 429 HIV-1 infected individuals, 95 of whom had histologically confirmed KS.
