This project investigates sterol and oxysterol content of murine brain and macrophages. Oxysterols are oxidised forms of cholesterol implicated in a wide array of biological functions. The compounds were analysed with LC-MS LTQ-Orbitrap high resolution system, which provides a highly sensitive and accurate tool for analysis of metabolites. Oxidized low density lipoproteins (ox-LDLs) are involved in the pathogenesis of atherosclerosis because of their cytotoxic and proapoptotic effect on vascular cells. However a dual effect of ox-LDL on endothelial cells (EC) has been demonstrated: they induce a proliferative effect at concentrations below 50 ?g/mL and a proapoptotic effect at concentrations above 50 ?g/mL. Hedgehog signaling is a pathway essential in embryonic development, adult stem cell maintenance, and is implicated in the formation and progression of cancer. Signaling in this pathway is triggered when the secreted protein Hedgehog binds to its membrane receptor, Patched. Patched normally inhibits the seven-spanner transmembrane protein Smoothened (Smo). Binding of Hedgehog inhibits Patched resulting in Smo derepression. Active Smo then triggers the activation of the cytoplasmic steps of the signaling pathway.

A mutation in ER membrane protein VAPB is responsible for causing a familial form of ALS (ALS8). The VAPB-P56S mutation causes protein aggregation and a nuclear envelope defect, where retrograde transport is disrupted. Over-expression of a FFAT peptide from OSBP1 reduces the size of VAPB-P56S aggregates and restores retrograde transport. A screen was performed on FFAT-motif containing ORPs to determine if any could rescue the mutant phenotype. ORP3 successfully reduced aggregate size and restored transport to the nuclear envelope.
