Abstract:
In the present work, I have investigated the role of oxytocin, a peptide hormone synthesised in the paraventricular and supraoptic (SON) nuclei of the hypothalamus and secreted from axon terminals in the posterior pituitary, in the initiation and maintenance of parturition in rats. Furthermore, I have studied afferent pathways involved in the secretion of oxytocin at the end of pregnancy, using immunocytochemical detection of Fos, the protein product of the immediate early gene c-fos. First, I have shown that delivery in rats, that has been disrupted by a systemic injection of morphine,

Under normal circumstances, prolactin regulates its own release via a short-loop negative feedback mechanism in which prolactin, secreted from lactotrophs situated within the anterior pituitary gland, stimulates dopaminergic neurons in the hypothalamus to release dopamine into portal blood circulation. Dopamine, in turn, inhibits lactotroph activity. A change in this regulation of prolactin secretion is seen during late pregnancy where tuberoinfundibular dopaminergic (TIDA) neurons no longer respond to elevated levels of placental lactogen (PL), a lactogen structurally and functionally similar

Abstract:
A delicate balance of steroid and gonadotrophic hormones is essential for intrafollicular oocyte maturation and successful fertilization and embryonic development. Previous studies have demonstrated that a superovulatory dose of pregnant mare serum gonadotrophin (PMSG) has excessive gonadotrophic activity and alters intrafol1icular steroid hormone levels. In a series of four experiments, the morphology of oocytes and embryos retrieved from immature rats, treated with either a low or high dose of PMSG, and mature, cycling rats was compared to determine whether a superovulatory dose of PMSG has

Hypertension and pregnancy are both accompanied by increases in sympathetic nerve activity. This has been attributed in part, to changes in the neurochemistry of the hypothalamic paraventricular nucleus (PVN). In hypertension, physiological studies have revealed that decreases in GABAergic inhibition and increases in glutamatergic excitation within the PVN contribute to this sympathoexcitation. In late-term pregnancy however, the sympathoexcitation appears to be mediated by decreases in GABAergic inhibition, with no glutamatergic contribution. Abstract:
Exercise and dietary carbohydrate restriction during pregnancy independently reduce maternal weight gain and offspring survival. It was hypothesized that the combined stress of exercise and dietary carbohydrate restriction would decrease offspring survival more than the independent effects. Within the exercise and sedentary groups pregnant rats were randomly assigned to be fed either 60%, 40%, or 20% dietary carbohydrate ad libitum. No statistical interactions were found between exercise and diet. Main effects were found for litter weight, maternal feed intake and weight gain, but not for litt

Abstract:
Since glucose is the principal substrate used during exercise and is also the main metabolic fuel for the developing embryo and fetus, exercising during pregnancy could induce a competition for fuel between fetus and exercising muscles, perturbing glucose homeostasis. To determine if exercise during pregnancy would predispose the fetus to increased risk, pregnant rats were randomly assigned to a low (4%), moderate (12%) or high (60%) carbohydrate diet, and either rested or exercised on a rodent treadmill at a moderate (15.5 m/min) or high (24.3 m/min) intensity from day 16-21 of gestation.
